吡哆胺对糖尿病大鼠视网膜晚期糖基化终产物及其受体的影响

摘    要:目的观察吡哆胺对糖尿病大鼠视网膜晚期糖基化终产物(AGEs)的含量和其受体(RAGE)表达的影响,探讨吡哆胺对糖尿病视网膜病的防治作用。方法 SD大鼠随机分为正常对照组(NC组)、糖尿病组(DM组)、吡哆胺治疗组(PM组)和糖尿病氨基胍治疗对照组(AG组),建立链脲佐菌素(STZ)糖尿病大鼠模型,每周测体质量和血糖。各组于治疗4周和12周取材,酶联免疫吸附(ELISA)法定量检测大鼠血清、视网膜中AGEs,12周时用免疫组织化学半定量检测视网膜RAGE的表达。结果 PM组、AG组与DM组比较,血糖差别无统计学意义;4周和12周时的PM组和AG组的大鼠血清和视网膜AGEs均低于DM组(P<0.05);12周时PM组和AG组视网膜RAGE表达低于DM组(P<0.01)。结论吡哆胺无明显降糖作用,但可通过减少AGEs堆积,降低RAGE表达,对糖尿病大鼠视网膜具有一定的保护作用。

Abstract:Objective To investigate the effect of pyridoxamine on advanced glycosylation end-products(AGEs) and their receptors in retina of diabetic rats.Methods Sprague-Dawley rats were randomly divided into four groups: normal control group(NC group),STZ-induced diabetes mellitus group(DM group),pyridoxamine group(PM group),and aminoguanidine group(AG group).Blood glucose level(BGL) and body weight were determined in different groups every week.ELISA was performed to quantify AGEs in serum and retina after 4 weeks and 12 weeks of treatment respectively.Immunohistochemistry was used to determine the expression of RAGE in retina of these rats after 12 weeks of treatment.Results Compared with those in DM group,blood glucose levels(BGL) in PM group and AG group were not different(P>0.05).4 and 12 weeks after treatment,compared with those in DM group,levels of AGEs in serum and retina in PM group and AG group decreased(P<0.05).12 weeks after treatment,compared with those in DM group,the expressions of RAGE in PM group and AG group increased significantly(P<0.01).Conclusion Pyridoxamine has no effect in decreasing blood glucose level,but has a certain role in protecting retina function by alleviating the accumulation of advanced glycosylation end-products and reducing the expression of RAGE in retina.
