剪切应力诱导成骨细胞钾离子通道开放的研究

摘    要:目的探索成骨细胞如何应答机械应力刺激,并将刺激信号转导进入细胞内,从而调节骨改建的分子机制。方法利用膜片钳及自行设计的流动小室系统对体外培养的单层Wister大鼠乳鼠成骨细胞在可控流体剪切应力作用下诱导细胞膜K 通道的开放进行了初步研究。结果剪切应力作用后,成骨细胞膜上K 电流明显增大。结论成骨细胞细胞膜上存在剪切应力敏感的K 通道,流动剪切应力可以通过影响成骨细胞膜上的K 通道的开放引起穿细胞的离子通透性的增加,进而引起细胞内Ca2 浓度变化,同时诱导细胞内钙调节机制的激活,这些变化都可以进一步引起细胞信号转导机制的激活。

Abstract:Objective To explore the mechanism of osteoblasts response to shear stress and transducing signal into cells and regulating bone reconstruction.Methods Using patch-clamp technique and parallel-plate flow chamber system,the cells membrane's K channel opening of rat's osteoblasts cultured in vitro was observed under controllable shear stress.Results Compared with nonshear stress,K current of osteoblast's membrane increased significantly in amplitude after shear stress stimulating.Conclusion The shear stress sensitive ion channel exists on the membrane of osteoblasts.Via the K channel opening of osteoblast's membrane,shear stress can induce the permeability increasing for ion penetration in cells,then results in changing of cell Ca2 conventration,and induces the activation of calcium modulation at the same time,all these changes activate signal transduction of osteoblasts further.
