穿透角膜移植术后房水前列腺素E2及蛋白质量浓度的动态变化

目的　通过对穿透角膜移植术后房水前列腺素E2 (PGE2 )和蛋白质量浓度的测定 ,探讨PGE2 对炎症反应和免疫反应的作用。方法　 42只健康新西兰白兔 ,按穿透角膜移植术的类型和处理方法不同 ,随机分为 6组 ,分别于术后第 10、2 0、3 0、40d抽取各组动物的房水 ,用放射免疫法检测PGE2 质量浓度 ,用紫外分光光度计法测定蛋白质量浓度。结果　自体移植组术后 10d房水PGE2 质量浓度高于对照组 ;同种异体移植组术后 10、2 0d房水PGE2 质量浓度高于对照组 ,1只兔眼于术后 2 0d出现排斥反应 ;异种移植组术后 10、2 0、3 0d房水PGE2 质量浓度均高于对照组 ,有 6只兔眼于观察期内出现明显排斥反应 ;异种移植双氯芬酸钠和地塞米松 (DFNa Dex)治疗组术后 10d房水PGE2 质量浓度低于异种移植Dex治疗组(P <0 0 5 )。房水蛋白质量浓度的变化情况与PGE2 相一致。结论　PGE2 与穿透角膜移植术后的炎症反应和免疫排斥反应密切相关。Dex可抑制异种穿透角膜移植术后的排斥反应。DFNa对Dex有协同作用 ,可减少Dex的用量。

Dynamic changes of prostaglandin E2 and protein level in rabbit aqueous humor following penetrating keratoplasty

ObjectiveTo investigate the effect of prostaglandin E 2(PGE 2) on inflammation and immune reaction by determining the levels of PGE 2 and protein in rabbit aqueous humor after the penetrating keratoplasty.MethodsForty-two healthy New Zealand white rabbits were divided into six groups randomly according to surgery type and treating method.The aqueous humor samples were collected on the postoperative day 10,20,30 and 40,respectively.Radioimmunoassay (RIA) and UV spectrophotometry were performed to detect the levels of PGE 2 and protein respectively.ResultsThe PGE 2 level in the aqueous humor was higher than that of normal group on the 10 thday after autokeratoplasty,on the 10 th and 20 th day after homokeratoplasty and on the 10 th,20 thand 30 thday after heterokeratoplasty.The PGE 2level in diclofenac sodium (DFNa) combined with dexamethasone (Dex) group was lower than that in Dex group on the 10 thday after heterokeratoplasty.The dynamic change of protein level was identical with PGE 2.ConclusionThe change of PGE 2in the aqueous humor is closely correlated with the inflammation reaction and the immune rejection.The Dex and DFNa can inhibit the immune rejection.
