林下参与栽培参扩增片段长度多态性指纹图谱鉴定

摘    要: 目的探讨林下参与栽培参DNA直接扩增片段长度多态性(directamplificationoflengthpolymorphisms,DALP)特征,建立林下参与栽培参直接扩增片段长度多态性指纹鉴定图谱。方法采用改进十六烷基三甲基溴化胺(CTAB)法提取林下参和栽培参基因组DNA,设计6条随机引物,优化直接扩增片段长度多态性体系,进行PCR扩增。结果采用改进的十六烷基三甲基溴化胺法从人参样品中提取到约为19kb左右的基因组DNA,其纯度在(1.80±0.23)之间,并获得林下参与栽培参的直接扩增片段长度多态性指纹图谱。结论获得的林下参直接扩增片段长度多态性图谱具有指纹特征,可以作为林下参与栽培参的特异鉴定方法。

Abstract:OBJECTIVE To explore the characteristics of DNA direct amplification of length polymorphisms (DALP) from wild ginsengs under forest and cultivated Panax ginseng. METHODS The CTAB method was modified to extract genomic DNA from wild ginsengs under forest and cultivated Panax ginseng. Six random primers were designed, and DALP system was optimized for PCR amplification. RESULTS Genomic DNAs of 19Kb were extracted by CTAB method from all the samples, and their purity was calculated to be (1.80±0.23). DALP fingerprints of wild ginsengs under forest and cultivated Panax ginseng were obtained. CONCLUSION The DALP fringerprint of wild ginsengs under forest has a specific characteristics, and can be used to identify wild ginsengs under forest and cultivated Panax ginseng.
