摘 要: | 目的分析新疆维吾尔族长寿老人肠道菌群多样性,比较肠道微生物种群的差异,以求找到肠内某些与健康有重要关系的有益菌。方法采用不依赖分离培养的16SrDNA的PCR-DGGE基因指纹技术,对新疆维吾尔族长寿老人肠道细菌进行检测。结果13份粪便标本肠道菌群总DNA均扩增出16S rDNA,大小约470bp;DGGE显示每种样品条带数目、亮度及位置不同,聚类分析显示不同个体粪便标本的细菌组成差异性非常大。结论新疆维吾尔族长寿老人肠道菌群具有多样性。基于16S rDNA的PCR-DGGE基因指纹分析可揭示肠道细菌组成的差异。

Abstract: | Objective To analyze the intestinal flora diversity among longevity aged people of Uygur nationality in Xin jiang, and compare species differences in intestinal microflora, so as to find the beneficial bacterium relating with the health. Methods Thirteen stool specimens were collected from longevity aged people, and the intestinal flora was detected by PCR-DGGE fingerprint technic. Results The total DNA of 13 specimens were amplified 16S rDNA, the size about 470 bp. DGGE showed that the number of band, location and brightness of each samples were different. The individuals clustering analysis showed that the variability of bacterial composition of each sample was great. Conclusion There are a variety of intestinal florea among longevity aged people of Uygur nationality in Xinjiang. The PCR-DGGE fingerprint based on the 16S rDNA gene can analyze the differences of composition of intestinal flora. |
