应用流式细胞术检测HIV-1感染者外周血p24、gp41和gp120抗原表达的研究

摘    要:目的分析HIV-1感染者外周血CD4^+T淋巴细胞p24、gp41和gp120抗原的表达状况,探讨流式细胞术用于检测HIV-1感染的可行性。方法应用三色流式细胞术,对192例HIV-1感染者和29例健康者外周血CD4^+T淋巴细胞表达的HIV-1 p24、gp41和gp120抗原进行检测。结果HIV-1 p24、gp41和gp120抗原阳性率HIV-1感染组与健康对照比较差异有统计学意义(P均〈0.01);CD4^+T细胞数〈200 cells/μl的HIV-1感染人群与CD4+T细胞数〉200 cells/μl的人群比较差异有统计学意义(P〈0.01或P〈0.05);静注吸毒途径感染人群与性途径感染人群比较差异有统计学意义(P〈0.01)。在HIV-1感染人群中,p24、gp41和gp120抗原阳性率四分位数25%75%区间范围分别为1.67%5.95%、1.61%8.12%和0.56%2.35%。结论流式细胞术简便、快速、敏感,可用于HIV-1感染检测。

Abstract:Objective To investigate the feasibility of detecting HIV-1 infection using flow cytometry(FCM) by analyzing the expression of p24,gp41 and gp120 antigens in peripheral blood CD4+ T lymphocytes from HIV-1 infected patients.Methods The expression of p24,gp41 and gp120 antigens in peripheral blood CD4+ T lymphocytes from 192 HIV-1-positive patients and 29 HIV-1-negative patients was detected by three-color flow cytometry.Results Results demonstrated that the frequency of p24,gp41 and gp120 in the HIV-1 positive group significantly differed from that in the HIV-1 negative group(P〈0.01).The expression frequency in infectious individuals with CD4+ T lymphocyte absolute counts of less than 200 cells/μl significantly differed from that in patients with CD4+ T lymphocyte absolute counts of more than 200 cells/μl(P〈0.01,P〈0.05 respectively).In addition,the expression frequency in patients infected by intravenous drug use differed significantly from that in patients infected by sexual c...更多ontact(P〈0.01).The frequency quartile of p24,gp41 and gp120 expression in HIV-1 infectious individuals was 1.67%-5.95%,1.61%-8.12%,and 0.56%-2.35% respectively.Conclusion Flow cytometry is a simple,quick,and sensitive method and has the potential to detect HIV-1 infection.
