依达拉奉对大鼠脑缺血再灌注后Bcl-2的影响

摘    要:目的本试验通过观察依达拉奉对Bcl-2的影响,探讨它的保护作用及作用机制。方法Wistar雌、雄性大鼠共42只,随机分为假手术组、盐水对照组及依达拉奉用药组,对照组及用药组再进一步分为6h、24h、48h,根据Koizumi线栓法建立大鼠左侧大脑中动脉缺血再灌注模型(MCAO)。用免疫组化方法检测Bcl-2的蛋白表达的变化。结果假手术组几乎无Bcl-2蛋白的表达,盐水组Bcl-2蛋白表达6h开始升高,于24h达到高峰并于48h开始下降。6h时,用药组Bcl-2蛋白表达较盐水组减少,但差异不明显(P〉0.05);24h时,用药组较盐水组明显减少(P〈0.01);48h时,用药组Bcl-2蛋白表达较盐水组减少,仍有明显差异(P〈0.05)。结论依达拉奉可抑制Bcl-2激活,达到脑保护作用。

Effect of edaravone on Bcl - 2 in rats with cerebral ischemia reperfusion.

Abstract:Objective The protective effect of edaravone was evaluated through observing expression of Bcl - 2. Methods Forty - two Wistar rats were randomly divided into 3 groups, sham operation group, saline control group and edaravone treatment group, saline control group and edaravone treatment group were divided into 6h,24h,48h. The left middle cerebral artery ischemia- reperfusion injury model (MCAO) was made by Koizumi suture - occluded method. The expression of Bcl - 2 was investigated by immuno - histochemistry technique. Results There was almost no expression of Bcl -2 in sham operation group. The expression of Bcl - 2 in saline control group increased in 6h and reached peak in 24 hours and continued to 48h. Compared with that in saline control group, the expression of Bcl -2 in edaravone treatment group obviously reduced ( P 〈 0.01 ). Conclusion Edaravone could reduce the expression of Bcl - 2 and protect the brain in rat.
