摘    要:目的研究bFGF对骨膜细胞的成骨性生长的影响。方法手术分离大鼠骨膜,用胰酶消化法分离骨膜细胞进行元代培养稳定传代后,选择第3~5代细胞随机分成两组;细胞同步化处理后,实验组用终浓度10ug/L的bFGF刺激细胞,对照组正常培养,72h后,用[3H]-TDR方法检测细胞的增殖状态,RT—PCR方法和Westernblot方法检测成骨细胞骨钙素基因的表达。结果10ug/L的bFGF处理细胞72h后,[3H]-TDR方法检测显示细胞DNA合成水平明显上调;而成骨细胞骨钙素基因在mRNA水平和蛋白水平的表达也明显上调。结论bFGF能刺激骨膜细胞的成骨性生长。

Abstract:Objective To study the effect of bFGF on periosteum cells. Methods The periosteum cells were cultured from femoral bone periosteum of the SD rats. The 3- 5 passage ceils were selected to the study and were randomly divided into control group and test group, control group was cultured by normal medium, bFGF of 10ug/L was added in test group. After 72h, two groups cells were detected through 3H -TDR method, and osteocalcin gene expression were detected by RT - PCR and Western blot. Results The cell DNA synthesis were obviously increased by 10ug/L of bFGF determined by 3H -TDR, and mRNA level and protein level of osteocalcin gene were also increased detected by RT- PCR and Western blot. Conclusion bFGF could stimulate the periosteum cells ossific proliferation.
