应用聚合酶链反应快速诊断巨细胞病毒感染
摘    要:用聚合酶链反应(PCR)技术检测学龄前儿童尿标本中的巨细胞病毒(HCMV),并和常规细胞培养(CCC)、早期抗原检测(DEA)两法进行了比较。结果表明,105份尿标本中有41份CCC出现HCMV特征性细胞病变(CPE),其中38份标本PCR检测为阳性,敏感性为92.7%;其余64份CCC阴性,有6份PCR检测阳性,特异性为90.6%。与DEA相比,PCR检测的特异性和敏感性分别为93.6%,95.2%;提示PCR检测临床标本中HCMV,不仅敏感特异,而且与CCC,DEA两法相比快速简便。

Abstract:In this study we used the polymerase reaction(PCR) protocol to detect the repetitive DNA sequences of human cytomegalovirus(HCMV) in urine samples from healthy infants.Out of 105 urine specimens tested,41 showed specific HCMV cytopathic effects(CPE) in conventional cell culture(CCC).Of these,38 were positive by the PCR detection(sentivity 92.7%).Of the remaining 64 urine specimens which were negative by the CCC,6 were positive by PCR (sensitivity 90.6%).In conclusion we consider that the PCR protocol is both sensitive and specific,It has several advantages over the CCC system and the most important of them is the rapidity with which it can achieve a diagnosis(in a few hours),while f1 disgnosis by CCC needs 15 days.
