参附注射液对急性坏死性胰腺炎肺损伤保护作用的研究

目的探讨急性胰腺炎时,血清TNF-α、IL-10水平和肺组织MDA含量、SOD活性的变化及参附注射液对它们的影响。方法健康SD大鼠30只分成3组:手术对照组(S组)、急性坏死性胰腺炎组(ANP组)、参附组(SF组),每组10只。采用5%牛磺胆酸钠诱导制备大鼠急性坏死性胰腺炎(ANP)模型。S组和ANP组于术前30分钟开始分别静脉泵注生理盐水10ml.kg-1.h-1,而SF组于术前30分钟开始静脉泵注参附注射液10mg·kg-1.h-1(1mg稀释为1ml)。4小时后活杀,每只大鼠取一叶左肺,光镜观察组织学改变。采用ELISA检测大鼠血清的TNF-α与IL-10水平;硫代巴比妥酸比色法测定丙二醛(MDA)含量;黄嘌呤氧化酶法测定过氧化物歧化酶(SOD)活性。结果MDA含量和TNF-α水平ANP组显著高于S组和SF组(p<0.01),且SF组MDA含量高于S组(p<0.05);S组和SF组SOD活性高于ANP组(p<0.05);IL-10水平ANP组显著低于S组(p<0.01),且低于SF组(p<0.05)。结论参附注射液能降低肺组织MDA含量和血清TNF-α水平,升高IL-10水平,保护SOD活性,对急性坏死性胰腺炎肺损伤有保护作用。

PROTECTIVE STUDY OF SHENFU INJECTION ON LUNG INJURY FOLLOWING ACUTE NECROSIS PANCREATITIS IN RATS

Objective To study the protective effect of Shenfu injection on 11Lung injury following acute necrosis pancreatitis. Methods 30 SD rats were randomized into three groups each consisting of 10 rats: Sham-operated control group(S group), acute necrosis pancreatitis group (ANP group), Shenfu group(SF group), ANP models were induced byretrograde injection of 5% sodium taurocholate in the pancreatic duct. TNF-α, IL-6 were detected by ELISA method. MDA was measured by thiobarbiturate colorimetry, and SOD activity was detected by xanthine oxidase technique. Results The content of MDA, TNF-α in ANP group was significantly higher than those in S and SFgroup (p<0.01),and MDA in SF group was higher than in S group (p<0.05).The SOD activity in S group and SF group were higher than that in ANP group(p<0.05). Meanwhile, IL-10 level in ANP group was significantly lower than in S group (p<0.01),and that was lower than in SF group (p<0.05). Conclusion Shenfu can decrease MDA content in lung tissue and TNF-α level in blood serum,increase IL-10 level in blood serum,protect SOD activity.Therefore,it can protect lung injury following acute necrosis pancreatitis.
