互隔交链孢霉毒素引起大鼠肝细胞DNA单链断裂的作用

摘    要:应用碱性洗脱荧光分析法对互隔交链孢霉产生的两种主要毒素——交链孢酚单甲醚(Alternariol Monomethyl Ether.AME)和交链孢酚(Alternariol.AOH)引起大鼠肝细胞DNA单链断裂效应进行了研究。实验结果表明:二者都能引起大鼠肝细胞DNA单链断裂,并有明显的剂量效应关系.DNA在滤膜上的存留分数明显低于溶剂对照组.具有统计学意义(P<0.01)。结果提示AME和AOH有致突变作用,二者可能是林县食管癌的病因之一。

Abstract:The alkaline elution and fluorometric DNA assay were adapted to the evaluation of DNA single-strand breaks in primary rat hepatocytes. The DNA damage was induced in vitro by the treatment withAlternariol monomethyl ether(AME)and Alternariol (AOH), which were the main mycotoxins of Al-ternaria alternata. The results showed that both AME and AOH could induce DNA single-strand breaksin primary rat hepaocytes and there existed dase--response association between the fractions of DNAremaining on the filter and the dose of AME and AOH. The fractions of DNA remaining on the filterin the test groupe were significantly lower than those in the solvent control (P<0. 01).
