加味清营颗粒对脂多糖诱导大鼠急性肺损伤模型炎性因子的影响

摘    要:目的 观察加味清营颗粒对急性肺损炎性因子的影响.方法 144只wistar大鼠随机分为空白对照组,模型组,加味清营颗粒低、中、高剂量组,地塞米松组,每组24只,连续灌胃6d,尾静脉注射LPS(5 mg/kg)后于1,3,5h不同时相处死大鼠.采用ELISA法检测肺泡灌洗液(BALF)中炎症介质白细胞介素(IL)-1β、肿瘤坏死因子(TNF)-α、IL-10的含量变化;用Bradford检测BALF蛋白含量;HE染色观察病理切片.结果 模型组各时相炎症因子均明显高于其他各组;与模型组比较,各治疗组均可显著降低的炎症介质IL-1β、TNF-α、IL-10的表达(P<0.05);与地塞米松组比较,加味清营颗粒低、中、高剂量组在1h、3h时相降低IL-1β、TNF-α的表达有统计学意义(P<0.05);与模型组相比较,各治疗组均可显著降低的BALF蛋白的表达(P<0.05);与地塞米松组比较,加味清营颗粒各时相剂量组在降低BALF蛋白的表达方面有统计学意义(P<0.05);肺组织HE染色病理结果示肺泡结构破坏或实变,肺泡隔增厚显著,毛细血管充血明显,肺泡腔内、细动脉周围和细支气管壁可见成堆炎症细胞浸润,而加味清营颗粒各剂量组肺组织病理均有较大程度的改善.结论 加味清营颗粒可降低急性肺损伤时炎症因子IL-1β、TNF-α、IL-10的释放,降低BALF中蛋白的含量,从而减轻肺部炎症细胞浸润,对损伤的肺组织起到修复保护的作用.

Effects of Modified Qingying Granule on Inflammatory Mediator Induced by Lipopolysaccharide in Rat Model with Acute Lung Injury

Abstract:Objective:To investigate the effect of Modified Qingying Granule on inflammatory mediator of acute lung injury.Methods:144 Wistar rats were randomly divided into blank control group,model group,Modified Qingying Granule low dose group,medium dose group and high dose group,and dexamethasone group (n =24).Continuous intragastric administration lasted 6 days.After tail vein injection of LPS (5 mg/kg),rats were executed at 1st,3rd,and 5th hour later.ELISA was used to detect the content changes of inflammatory mediators such as IL-1β,TNF-α and IL-10 in bronchoalveolar lavage fluid (BALF).BALF protein was examined by Bradford.The pathological section was made into HE stainining.Results:Inflammatory cytokines at different time points of model group were significantly higher than those in the other groups.Compared with model group,expressions of inflammatory mediators of treatment groups decreased (P〈 0.05).Compared with dexamethasone group,Modified Qingying Granule low,midium and high dose groups at 1 h and 3 th hour had statistical significance in reducing IL-1β and TNF-α expressions (P〈 0.05).Compared with model group,expression of BALF protein in treatment groups significantly decreased (P〈 0.05).Compared with dexamethasone group,Modified Qingying Granule had statistical significance in reducing BALF protein expression (P〈0.05).Lung tissue HE staining pathological results revealed damaged or changed alveolar structure,thickened alveolar septa,capillary congestion,and visible piles of inflammatory cell infiltration in alveolar space,around arteriole and in bronchiolar wall.Modified Qingying Granule groups had a greater degree of improvement in lung tissue pathology.Conclusion:Modified Qingying Granule can reduce the expressions of inflammatory mediators such as IL-1β and TNF-α,increase expression of IL-10,reduce the content of bronchoalveolar lavage fluid protein,so as to reduce the pulmonary inflammatory cell infiltration and to repair the protective function of damag
