附子干姜配伍对阿霉素致大鼠慢性心衰的治疗作用

摘    要:目的 观察附子于姜配伍治疗阿霉素(ADR)致慢性心衰的作用,并探讨其作用机理.方法 采用盐酸ADR腹腔注射方法,建立慢性心力衰竭大鼠模型,将SD大鼠随机分为6组:对照组,模型组,附子高、低剂量组(3.3,1.65 g/kg),附子干姜(1∶1)高、低剂量组(6.7,3.3 g/kg).采用酶联免疫吸附法(ELISA)检测大鼠血清中去甲肾上腺素(NE)、内皮素-1(ET-1)和心脏组织中Na^+-K^+-ATPase的活性,运用蛋白免疫印迹杂交法(Western Blot)检测心脏组织蛋白Junctophilin-2 (JP2)的蛋白表达.结果 与模型组比较,附子和附子干姜配伍血清中NE及ET-1含量显著降低,心脏组织中Na^+-K^+-ATPase含量及JP2的蛋白表达显著升高,其中附子干姜(1∶1)高剂量组作用更显著(P<0.01).结论 附子与附子干姜(1∶1)配伍对ADR致慢性心衰的作用,可能与其增加心脏组织Na^+-K^+-ATPase活性,上调Junctophilin-2 (JP2)的蛋白表达有关.

Study on the Compatibility of Aconite and Dried Ginger Treating Rats with Adriamycin-induced Chronic Heart Failure

Abstract:Objective: To study the compatibility of aconite and dried ginger treating rats with adriamycin-in- duced chronic heart failure and to explore its mechanism. Methods: Hydrochloride adriamycin injection was ap- plied to establish an chronic heart failure animal model, SD rats were randomly divided into 6 groups: normal group, model group, aconite high and low dose groups (3.3,1,65 g/kg), and aconite and dried ginger( 1:1 ) high and low dose groups(6.7,3.3 g/kg). The content of NE and ET-1 in rat serum and activity of Na^+-K^+-ATPase in car- diac tissue were detected by ELISA.JP2 protein expression in heart tissue was detected by western blot. Results: Compared with model group,the levels of NE and ET-1 in serum significantly decreased while the activity of Na^+-K^+-ATPase and expression of Junctophilin-2 (JP2) protein in cardiac tissue significantly increased. Compati- bility of aconite and dried ginger (1:1) had more significant effect on the high dose group. Conclusion: Compati- bility of aconite and dried ginger treating adriamycin--induced chronic heart failure may relate to increasing activ- ity of Na^+-K^+-ATPase and enhancing expression of Junctophilin-2(JP2) protein.
