HPLC法测定威麦宁胶囊中原花青素B2和表儿茶素的含量

目的:建立威麦宁胶囊中原花青素B2、表儿茶素的HPLC测定方法。方法:采用高效液相色谱法测定原花青素B2、表儿茶素的含量,其标准曲线、精密度、重复性、加样回收率均符合要求。色谱柱:Zorbax SB-C18柱,流动相为乙腈-0.04%磷酸水溶液,梯度洗脱,检测波长为280nm,柱温30℃,流速1.0 mL.min-1,进样量10μL。结果:原花青素B2、表儿茶素的标准曲线分别为A原花青素B2=7047.3C-1066(r=0.9998),A表儿茶素=7321.9C-601.43(r=0.9999),加样回收率分别为93.56%(RSD 4.36%)、99.34%(RSD 4.14%),3个批次威麦宁胶囊中原花青素B2、表儿茶素的平均含量分别为1.44mg/粒、0.96mg/粒。结论:威麦宁胶囊3个批次中2种主要成分的含量相近。该方法简便、准确、重复性好,可为威麦宁胶囊提供质量控制方法。

Determination of Procyanidin B2 and Epicatechin in Weimaining Capsules by HPLC

Objective： To establish an HPLC method for the determination of procyanidin B2 an epicateehin in Weimaining capsules. Methods： An HPLC method was established with its standard curves, precisions, repealabilities and recoveries met to the requirements. The determination was performed on a Zorbax SB-C18 cohnnn. The mobile phase was eousisted of aeetonitrile-0.04% phosphoric acid （gradient chillon）. The delection wavelength was set al 280 mn and the column temperature was 30℃. The flow rate was 1.0 mL ·min^-1 The injection volumn was 10 /M,. Results： The standard curves were Aprocyaindin B2 = 7047.3C-1066 （r=0.9998） and Aepicatechin=7321.9C-601.43 （r=0.9999） with average recoveries at 93.56% （RSD 4.36%） and 99.34% （RSD 4.14%）, respectively. The average content of procyanidin B2 and epicateehin in 3 batches of Weimaining Capsules were 1.44 mg and 0.96 mg per capsule, respectively. Conclusion： The conlents of procyanidin B, and epieatechin in 3 halehes of Weimaining Capsule are similar. The method is simple, accurale and reproducible for quality control of Weimaining capsules.
