辛基酚和三羟异黄酮对大鼠乳腺癌发生及细胞增殖影响的研究

摘    要:目的研究辛基酚和三羟异黄酮在体内、外条件下对乳腺癌发生、增殖的影响.方法将20 d龄雌性SD大鼠92只完全随机分为阴性对照组(8只)、阳性对照组(21只)、三羟异黄酮处理组(GEN组,21只)、辛基酚处理组(OP组,21只)及GEN、OP联合处理组(GP组,21只),采用二甲荃苯蒽(7,12-dimethyl benz[a] anthracence, DMBA)诱发大鼠乳腺癌模型,观察OP、GEN干预对乳腺癌发生的影响.同时,体外培养MCF-7细胞,观察OP、GEN对细胞增殖的影响,运用MTT比色法检测各组乳腺癌细胞的增殖功能,用免疫荧光细胞化学法检测细胞中增殖细胞核抗原(proliferation cell nuclear antigen, PCNA)的表达.结果与对照组相比较,GEN大鼠肿瘤体积显著降低(P<0.05),肿瘤数目降低更为显著(P<0.01);OP组肿瘤体积显著降低(P<0.05);GP组肿瘤数目和肿瘤体积均显著降低(P<0.05).与对照组相比较,GEN组培养72 h后MCF-7细胞增殖及PCNA表达均显著降低(P<0.01);OP组细胞增殖能力显著增强(P<0.01),PCNA表达至72 h显著上升(P<0.05);GP组MCF-7细胞的增殖及PCNA表达均显著降低(P<0.01).结论辛基酚、三羟异黄酮能分别促进和抑制DMBA诱导的大鼠乳腺癌发生及体外MCF-7细胞的增殖;且二者共同作用时,后者能抑制前者的促进作用.

Abstract:Objective To study the effects of octylphenol (OP) and genistein (GEN) on the incidence and proliferation of mammary tumor in vitro and in vivo. Methods A total of 84 young female SD rats were randomly divided into four groups: control, GEN, OP, and GP groups. Rat breast cancer was established by gastric irrigation of DMBA at the dose of 100 mg/kg (body weight), and then the effects of OP and GEN intervention on the incidence of breast cancer were observed. Meanwhile, we also observed the effects of OP and GEN intervention on the proliferation of MCF-7 cells in vitro by MTT and immunohistochemical staining of the proliferating cell nuclear antigen (PCNA). Results Compared with the positive control group, the tumor volumes and number in GEN group decreased significantly (P<0.05 and P<0.01, respectively). In OP group, the tumor volumes decreased significantly (P<0.05). In GP group, the tumor number and volumes decreased significantly (P<0.05). Compared with that in the control group, the proliferation of MCF-7 cells in OP group increased significantly (P<0.01), but in GEN and GP groups, the proliferation and expression of PCNA decreased significantly (P<0.01). Conclusion OP and GEN can respectively promote and inhibit the incidence of breast cancer induced by DMBA and the proliferation of MCF-7 cells. GEN can antagonize the effect of OP when they are used together.
