摘    要:目的探讨乌鸡白凤丸有效成分(BFP)对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱致C57BL小鼠黑质神经元凋亡的保护作用及其可能机制。方法C57BL小鼠连续5d皮下注射MPTP(30mg/kg)制备帕金森病小鼠模型。给予BFP预处理后,用酪氨酸羟化酶(TH)免疫组织化学法观察黑质神经元的变化;同时应用免疫组织化学法检测Bax蛋白表达; 应用高效液相-电化学方法检测纹状体(Str)多巴胺(DA)及其代谢产物二羟基苯乙酸(DOPAC)和高香草酸(HVA)的含量变化。结果BFP预处理能使黑质致密带(SNc)TH免疫阳性神经元数目较MPTP组明显增多,而Bax蛋白表达较MPTP组明显减少;可使Str区DA及其代谢产物DOPAC、HVA含量明显增加。结论BFP有效成分可以拮抗神经毒性物质MPTP对C57BL小鼠黑质多巴胺能神经元的损伤,其作用机制可能与其抗凋亡作用有关,Bax表达的降低可能是BFP抗凋亡作用的途径之一。

Abstract:ObjectiveTo explore the neuroprotective effect of Bak Foong Pill(BFP) extract on the apoptosis of substantia nigra neurons induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) in C57BL mice and its possible mechanism. MethodsIn the control group,C57BL mice received MPTP(30 mg/kg) subcutaneous injection for five days to establish the PD model.In the pretreatment group,Bak Foong Pill extracts were given for 14 days prior to MPTP.Immunostaining of tyrosine hydroxythase(TH) was used to observe the damage of dopamine(DA) neurons,and of Bax to detect the protein levels of Bax.High performance liquid chromatography-electrochemical detection(HPLC-ECD) was used to investigate the contents of DA and its metabolites dihydroxy-phenylacetic acid(DOPAC) and homovanillic(HVA) in the Str.ResultsCompared with the control group,neurons in the substantia nigra zona compacta of the pretreatment group markedly increased the immunoexpression of TH and reduced the expression of Bax.Contents of DA and its metabolites DOPAC,HVA were also increased.ConclusionBFP extracts showed protective effect on dopaminergic neurons against MPTP-induced neurotoxicity in the PD model mice.The neuroprotective effect might be attributed to anti-apoptosis manifested by decreased expression of Bax.
