美洲大蠊前处理工艺及其体外抗肿瘤活性研究

目的:研究美洲大蠊前处理工艺对蛋白质含量及其体外抗肿瘤的影响。方法:通过沸水、低温两种方式分别将美洲大蠊烫杀、冻杀,用传统干燥、真空干燥、冷冻干燥三种方式干燥美洲大蠊,以得率、含水量、醇浸出物含量、蛋白质含量为理化指标,以体外抗肿瘤实验为活性指标,选出最佳的前处理工艺。结果:蛋白质得率高低为冷冻干燥真空干燥传统干燥。含水量高低为真空干燥传统干燥冷冻干燥。醇浸出物含量高低为真空干燥传统干燥冷冻干燥。蛋白质含量测定高低为真空干燥冷冻干燥传统干燥。体外细胞实验:真空干燥对A549、HeLa、MCF-7、SGC-7901、Bel-7402五种细胞均具有较强抑制作用,其次为冷冻干燥,传统干燥效果最弱。结论:本实验通过比较三种前处理工艺对美洲大蠊中蛋白质含量及其体外抗肿瘤的影响,确定以真空干燥为美洲大蠊前处理工艺,为美洲大蠊的进一步开发提供依据。

Study on the Pre-treatment Process and Its Antitumor Activity in vitro of Periplaneta Americana

Objective:To study the effect of pretreatment technology on protein content and antitumor activity in vitro of Periplaneta americana. Methods:Periplaneta americana was sacrificed in two ways by boiling water and low temperature,and was further dried using traditional drying,vacuum drying and freeze drying.The optimized pretreatment process was determined with the yield,water content,content of alcohol extract and protein content as physical and chemical indexes,and antitumor effect in vitro as the activity index.Results:The yield of Periplaneta Americana in descending sequence is as follows:freeze drying > vacuum drying > traditional drying;water content:vacuum drying > traditional drying > freeze-drying;the content of alcohol extract:vacuum drying > traditional drying > freeze-drying;protein content:vacuum drying > freeze drying > traditional drying,and cell experiment in vitro showed that vacuum drying had the strongest inhibitory effect on A549,HeLa,MCF-7,SGC-7901 and Bel-7402 cells,followed by freeze drying,and the traditional drying effect is the weakest. Conclusion:By comparing the effects of three preprocessing techniques on the protein content in Periplaneta americana and its antitumor effect in vitro,vacuum drying was determined as a pretreatment process for Periplaneta americana. This study would provide a reference for the further development of Periplaneta americana.
