十枣汤联合顺铂干预恶性腹水的作用机制研究

摘    要:目的:探讨十枣汤联合顺铂干预恶性腹水的作用机制。方法:将50只昆明种小鼠随机分为空白组、模型组、顺铂组、中药组、中药+顺铂组各10只。除空白组,其他各组复制艾氏腹水瘤模型,建模24 h后开始给药,给药10 d后处死小鼠,记录小鼠体质量,测量腹水量及脾和胸腺的质量,酶联免疫吸附测定(ELISA)法检测小鼠血清中白细胞介素2(IL-2)、干扰素γ(IFN-γ)、肿瘤坏死因子α(TNF-α)、血管内皮生长因子(VEGF)含量。结果:给药干预后,与模型组比较,除空白组外,各治疗组腹水明显减少(P<0.01),中药+顺铂组效果最佳,IL-2、IFN-γ含量显著升高(P<0.01),TNF-α、VEGF含量显著降低(P<0.01)。结论:十枣汤与顺铂联用能够明显抑制恶性腹水,其机制可能与升高IL-2、IFN-γ含量及降低TNF-α、VEGF含量有关。

Study on the Mechanism of Shizao Decoction Combined with Cisplatin in Intervention of Malignant Ascites

Abstract:Objective:To investigate the mechanism of Shizao Decoction combined with cisplatin in the treatment of malignant ascites. Methods:Fifty Kunming mice were randomly divided into blank group,model group,cisplatin group,Chinese medicinal group,and Chinese medicinal + cisplatin group. Except the blank group,in all groups the Ehrlich ascites tumor model was established,and the drug was administered 24 hours after modeling. The mice were sacrificed 10 days after the administration,and the weight was recorded. The amount of ascites and the weight of the spleen and thymus were measured. The serum levels of interleukin-2(IL-2),interferon γ(IFN-γ),tumor necrosis factor α(TNF-α),and vascular endothelial growth factor(VEGF) were detected in mouse serum using enzyme linked immunosorbent assay(ELISA). Results:After intervention,compared with the model group,the ascites of each treatment group was significantly decreased(P <0.01),and the effect in the Chinese medicinal + cisplatin group was the best,IL-2 and IFN-γ levels were significantly increased(P<0.01),TNF-α and VEGF levels were significantly decreased(P<0.01). Conclusion:The combination of Shizao Decoction and cisplatin can significantly inhibit the development of malignant ascites. The mechanism may be related to the increase of IL-2 and IFN-γlevels and the decrease of TNF-α and VEGF levels.
