山楂黄酮对微波辐射所致Wistar大鼠脑损伤的治疗作用

摘    要:目的:探讨山楂黄酮对微波辐射所致Wistar大鼠脑损伤的治疗作用,阐明山楂黄酮对微波产生的非热效应和热效应的治疗效果。方法:36只Wistar大鼠随机分为正常对照组、辐射模型组和辐射给药组。辐射模型组和辐射给药组大鼠给予200 mW.cm-2的微波辐照5 min,辐射给药组大鼠于辐射后给予40 mg.kg-1 的山楂黄酮,每日1次,连续14 d。在开始给药后的10、11、12、13和14 d采用Morris水迷宫检测大鼠学习与记忆能力的变化,停药后隔日处死大鼠取大脑,采用光学显微镜观察脑组织结构的变化。结果:与对照组比较,200 mW.cm-2微波辐照后辐射模型组和辐射给药组大鼠Morris水迷宫目标象限时间、平台停留时间、经过平台次数和经过有效区次数均明显下降(P<0.05);与辐射模型组比较,辐射给药组大鼠上述数据均升高(P<0.05)。200 mW.cm-2微波辐照后大鼠海马与小脑神经元胞核固缩、深染,辐射给药组大鼠海马神经元损伤有所减轻,呈恢复状态,小脑神经元和血管无明显变化。结论: 200mW.cm-2微波辐照后可损伤大鼠学习记忆能力和脑组织结构,山楂黄酮对微波辐射所致损伤有一定的治疗作用。

Abstract:Objective To study the curative effect of Hawthorn flavones on brain damage in Wistar rats induced by microwave radiation and to clarify the treatment effect of Hawthorn flavones on non-heat effect and heat effect induced by microwave radiation.Methods 36 Wistar rats were randomly divided into normal control group,radiation model group and radiation administration group.The rats in the last two groups were given 200 mW·cm-2 microwave radiation for 5 min.After microwave radiation,the rats in radiation administration group were given 40 mg·kg-1 Hawthorn flavones once a day,the rats in the other two groups were rendered the same amount of saline by the same way.The whole process lasted for 14 d.Then the learning and memory capacities of the rats in 10,11,12,13 and 14 d respectively were detected by Morris Water Maze.The other day after stopping given the Hawthorn flavones,the rats were killed and the brains were removed,then the changes of the brain structures were observed by optical microscope.Results Compared with normal control group,the target quadrant time,platform residence time,times of crossing platform and times of crossing coverage area of rats in radiation model group and radiation administration group were decreased after 200 mW·cm-2 microwave radiation(P<0.05);meanwhile the indexes mentioned above in radiation administration group were higher than those in radiation model group(P<0.05).After 200 mW·cm-2 microwave radiation,the rat hippocampus and cerebellum neurons appeared nuclear pyknosis and deep dyeing,and in radiation administration group the injury of rat hippocampus neurons was decreased,and restored,without significant changes in cerebellum neurons and blood vessels.Conclusion The learning and memory capacities and the brain structure of the rats can be damaged by microwave radiation with dosage of 200 mW·cm-2 and Hawthorn flavones has curative effect on brain damage of rats induced by microwave radiation.
