摘    要:目的:探寻与G-CSF受体胞内区相互作用的下游蛋白因子,揭示G-CSF受体信号转导通路的可能机制。方法:采用敏感性较高的酵母双杂交体系,筛选小鼠胎肝文库,并通过体内双杂交实验进行验证。结果:获得了11种阳性基因片段,对其中铁蛋白重链片段在体内验证了相互作用。结论:提示在小鼠胎肝中G-CSF受体作为胞内信号转导的起始点,与众多胞浆内蛋白因子相结合,对于揭示G-CSF受体信号转导可能存在的各种通路起到重要的提示作用。

Abstract:Objective:To explore the protein factors that may interact with G_CSF receptor in the signal transduction pathways.Methods:The highly sensitive protein_protein interaction system--yeast two_hybrid system was applied to screen the mouse fetal liver library. The interaction of GST_ferritin with His_GCSFR in vitro was examined with mammalian two_hybrid expriment.Results:Eleven positive gene fragments were obtained,and mammalian two_hybrid experiment confirmed that ferritin heavy chain interacted with intracellular G_CSF receptor.Conclusions:It is the first time to find that in mouse fetal liver tissue, G_CSF receptor can interact with many protein factors to activate some signaling pathways. This result may be important for understanding the relationship between protein factors and G_CSF receptor.
