小蘗碱诱导人宫颈癌Hela细胞株凋亡的作用

目的:探讨中药黄连素对子宫颈癌Hela细胞株的增殖抑制和诱导凋亡的作用。方法:以不同浓度的黄连素(10、20、40、80μmol/L),分12、24、48、72h四个时间点处理Hela细胞,光镜观察细胞形态变化;用四甲基偶氮唑蓝(MTT)法测定细胞增殖抑制率;用透射电子显微镜及琼脂糖凝胶电泳检测凋亡发生情况。结果:黄连素可抑制Hela细胞增殖,作用呈明显的时效和量效关系,差异有统计学意义(P<0.01);黄连素可诱导Hela细胞凋亡,透射电子显微镜下可见凋亡细胞;琼脂糖凝胶电泳出现细胞凋亡典型的DNA"梯状"条带。结论:黄连素体外对Hela细胞具有增殖抑制作用和促进凋亡作用。

The apoptosis of human cervical cancer Hela cell line induced by berberine

Objective:To explore the effect of berberine in inhibiting the proliferation of cervical cancer Hela cell line and in ducing apoptosis.Methods:Hela cells were treated with various concentrations of Berberine(10 μmol/L,20 μmol/L,40 μmol/L,80 μmol/L) for different periods(12 hours,24 hours,48 hours,72 hours),the changes of cell morphology were observed by light microscope;the inhibition rate of cell proliferation was detected by MTT method;the occurrence of apoptosis was detected by electron microscope and agarose gel electrophoresis.Results:Berberine inhibited Hela cell proliferation,showing a obvious dose-and time-dependent manner,there was significant difference(P<0.01).Berberine induced Hela cells apoptosis,which could be observed under electron microscope;agarose gel electrophoresis showed a characteristic DNA ladder of cell apoptosis.Conclusion:Berberine can inhibit cell proliferation and promote cell apoptosis of human Hela cells.
