UNC5B在人早孕胎盘微血管内皮细胞中的表达及意义

目的:探讨神经轴突导向因子Netrin-1受体在人早孕胎盘微血管内皮细胞(HPMECs)中的表达及意义。方法:采用Percoll密度梯度离心法分离HPMECs,并进行鉴定。实时定量PCR技术检测HPMECs中Netrin-1的6种受体(UNC5A、UNC5B、UNC5C、UNC5D、DCC和neogenin)mRNA的表达;应用RNA干扰技术检测Netrin-1受体对HPMECs凋亡的影响。结果:①成功分离HPMECs,免疫荧光鉴定结果显示Ⅷ因子相关抗原和CD34呈阳性。②实时定量PCR结果显示,Ne-trin-1的6种受体中,仅检测到UNC5BmRNA在HPMECs中存在表达。③转染siRNA-UNC5B质粒后,HPMECs凋亡率较对照组明显下降(P<0.05)。结论:Netrin-1受体UNC5B在HPMECs中表达且能促进HPMECs的凋亡,提示其在胎盘血管形成过程中有重要作用。

Expression of UNC5B in human placental microvascular endothelial cells of early pregnancy and its significance

Objective:To explore the expression of neural axon guidance factor Netrin-1 in human placental microvascular endothelial cells(HPMECs)of early pregnancy and its significance.Methods:Percoll density gradient centrifugation was used to isolate HPMECs and identify;real-time quantitative PCR was used to detect the expression levels of six receptors mRNA of Netrin-1 in HPMECs(NC5A,UNC5B,UNC5C,UNC5D,DCC and neogenin);the effects of Netrin-1 receptors on apoptosis of HPMECs were detected by RNA interference technique.Results:HPMECs were isolated successfully,immunofluorescence showed that factor Ⅷ-related antigen and CD34 were both positive;real-time quantitative PCR indicated that only UNC5B mRNA expressed in HPMECs among six receptors of Netrin-1;compared with control group,the apoptosis rate of HPMECs decreased after transfection by siRNA-UNC5B plasmid(P<0.05).Conclusion:Netrin-1 receptor UNC5B expresses in HPMECs and promotes the apoptosis of HPMECs,which indicates that UNC5B plays an important role in placental angiogenesis.
