摘    要:目的:探讨RNA干扰沉默EZH2基因对人卵巢癌细胞系A2780增殖和迁移的影响。方法:真核表达载体介导靶向沉默EZH2 shRNA的重组质粒转染A2780细胞,Real-time RT-PCR检测EZH2基因的沉默效果,MTT法检测细胞增殖,流式细胞仪检测细胞周期,Transwell小室检测细胞迁移。结果:EZH2shRNA转染明显下调EZH2的表达(P0.001)。重组质粒稳定转染至A2780细胞后,细胞增殖速度明显下降(P0.05),G0/G1期细胞增加(P0.01),体外迁移能力下降(P0.01),差异均有统计学意义。结论:EZH2基因沉默能有效抑制卵巢癌细胞的增殖和迁移能力,为卵巢癌的表观遗传学治疗提供新靶点。

Inhibition effect of cell proliferation and migration by RNA interference silencing EZH2 gene in ovarian cancer cell line A2780
Abstract:Objective:To investigate the effect of EZH2 knockdown on cell proliferation and migration in ovarian cancer cell line A2780 by RNA interference.Methods:After the shRNA plasmid vector targeting EZH2 was transferred into A2780 cells,the mRNA expression of EZH2 was determined by Real-time RT-PCR,cell proliferation and migration capability were examined by MTT assays and Transwell migration assay,cell cycle was analyzed by Flow Cytometry.Results:EZH2 shRNA transference down regulated EZH2 mRNA successfully(P<0.001).EZH2 knockdown caused G0/G1 cell cycle arrest,suppressed cell proliferation capability as well as cell migration capability of A2780 cells(P<0.05).Conclusion:RNA interference silencing EZH2 gene can significantly inhibit cell proliferation and migration of A2780 cells,thus provide a novel target for epigenetic therapy of ovarian cancer.
