应用PCR技术研究白纹伊蚊的吸血习性

摘    要:目的应用PCR技术鉴定登革热传播媒介白纹伊蚊胃血来源,研究白纹伊蚊的吸血习性。方法在实验室条件下,将6种动物(鸡、猫、狗、鸽、野鼠、小鼠)分别放人6个小玻璃房,同时,将6种动物放入一个大玻璃房,然后放入已经饥饿18h的白纹伊蚊雌蚊,3h后采集吸血蚊,应用PCR技术鉴定白纹伊蚊胃血来源。现场调查则通过紫外诱蚊灯和BG.SentinelTrap采集吸血蚊,利用实验室建立的PCR体系鉴定白纹伊蚊胃血来源。结果在不同空间内,白纹伊蚊对野鼠的平均吸血率最高,为44.45%,其余依次为鸡、猫、小鼠、狗、鸽(38.27%、36.53%、35.70%、34.94%、24.75%)。在同一空间内,白纹伊蚊对鸡、小鼠、猫、野鼠、狗、鸽的吸血率依次为12.36%、7.09%、6.41%、4.81%、2.52%、1.60%。在现场收集到的饱血白纹伊蚊共13只,其中吸狗血的有7只,吸人血的白纹伊蚊有2只,多重吸血(sb鼠和猫)的白纹伊蚊有1只。结论PCR技术可鉴定白蚊伊蚊胃血来源,结果稳定可靠;白纹伊蚊不仅嗜吸人血,也可兼吸多种常见的动物血。

Abstract:Objective To develop a specific PCR method for the identification of the source of A edes albopictus, and to determine the blood-sucking habit of Aedes albopictus. Methods Under the laboratory conditions, six different animals (chicken, cat, dog, pigeon,voles, mice) were separately placed into six small glass rooms. Another same six animals were placed in a large glass room. These animals were then exposed to starved female mosquitoes of Aedes albopictus. Mosquitoes were collected 3 h later. In the field survey, samples were collected through the UV Ovitrap lights and BG- Sentinel Trap. The source of blood sucked by the mosquitoes was determined by PCR. Results When the animals were housed in separated glass room, the blood-sucking rate was in the order of vole (44.45%), chicken (38.27%), cat (36.53%), mouse (35.70%), dog (34.94%) and pigeon (24.75%). When all of the animals were housed in the same glass room, the blood-sucking rate was in the order of chicken (12.36%), mouse (7.09%), cat (6.41%), vole (4.81%), dog (2.52%), and pigeon (1.60%). Among the 13 samples obtained from the field survey, 7 blood samples were found from dog, 2 from human, and 1 sample showed a mixture of mice and cat blood. Conelusion The developed PCR method can identify the source of blood meals and the result is reliable and reproducible. Aedes albopictus appears to feed on not only human blood, but blood from other animals.
