腹腔注射全氟化碳对急性肺损伤大鼠肿瘤坏死因子水平的影响

目的观察腹腔注射全氟化碳对急性肺损伤大鼠肿瘤坏死因子(TNF-α)水平的影响。方法62只雄性Wista大鼠随机分为脂多糖(LPS)组、全氟化碳组(PFC组)、正常对照组(NC组)。PFC组于实验前48 h腹腔注射C8F18 15 ml.kg-1,实验日LPS组、PFC组静脉注射内毒素7 mg.kg-1,分别于2、46、h后处死,比较各组肺脏病理、血清和肺泡灌洗液中TNF-α水平。结果PFC组注射LPS后6 h肺泡灌洗液中TNF-α为9.60±0.61 pg.ml-1,明显低于LPS组(10.750±1.095 pg.ml-1)(P<0.05),其余时间点及血清中TNF-α水平两组无显著差异(P>0.05)。PFC组与LPS组病理评分无明显差异(P>0.05)。结论腹腔注射C8F18原液可以部分降低肺内的TNF-α水平,这可能可以减轻肺部的炎性损伤。

Effects of perfluorocarbon intraperitoneal injection on tumor necrosis factor-alpha levels in rats with acute lung injury

Objective To evaluate the effects of perfluorocarbon(PFC) intraperitoneal injection on tumor necrosis factor-alpha(TNF-α)levels in rats with acute lung injury(ALI).Methods Sixty Wistar rats were divided into normal control-group,LPS-group and PFC-group.PFC-group rats were treated with intraperitoneal injection of C8F18 15 ml·kg~(-1) at 48 h before the time of injury,then LPSgroup and PFC-group were injected with 7 ml·kg~(-1)LPS.Pathological grade,TNF-αlevel in serum and BALF at 2 h,4 h and 6 h were assessed respectively.Results The level of TNF-αin BALF in PFC-group at 6 h was lower than LPS-group(9.60±0.61 pg·ml~(-1) vs 10.75±1.09 pg·ml~(-1),P<0.05).There was no difference of pathological grade between two groups.Conclusion The intraperitoneal injection of PFC can reduce levels of TNF-αin lung,which may attenuate inflammation of ALI rat.
