高压氧下甲氨喋呤透过血脑屏障的观察(论著)

目的：观察高压氧（ｈｙｐｅｒｂａｒｉｃｏｘｙｇｅｎ，ＨＢＯ）对甲氨喋呤（ｍｅｔｈｏｔｒｅｘａｔｅ，ＭＴＸ）透过血—脑屏障（ｂｌｏｏｄ－ｂｒａｉｎｂａｒｒｉｅｒ，ＢＢＢ）的影响。方法：新西兰大白兔１２只，随机分为ＨＢＯ组（６只）、对照组（６只）。均予耳缘静脉注射ＭＴＸ１００ｍｇ／ｋｇ。静注药物后２小时采取血浆、脑脊液（ＣＳＦ）各２ｍｌ立即送检。用高效液相法测定ＭＴＸ的浓度，仪器用日本岛津ＨＰＬＣ（ＬＣ－６Ａ）。结果：静注药物１．５小时后，ＨＢＯ组ＣＳＦ中ＭＴＸ含量是血浆中的２．６７％，而对照组为１．２４％，两者经ｔ检验具有显著性差异（Ｐ＜０．０１）。结论：本实验发现ＨＢＯ下ＭＴＸ透过ＢＢＢ进入ＣＳＦ的量明显增多（Ｐ＜０．０１），这一现象有重要临床意义，因对于体肿瘤合并脑肿瘤的患者如作ＨＢＯ可以有增敏化疗效果，其ＣＳＦ中的ＭＴＸ含量会比不作ＨＢＯ时显著增多。至于是否还需要作鞘内注射，应结合临床情况考虑

Observation of the permeability of methotrexate through blood brain barrier under hyperbaric oxygen

Aim:To observe the effect of hyperbaric oxygen(HBO)on the permeability of methotrexate(MTX)through the blood brain barrier(BBB).Methods:12 rabbits were divided into 2 equal groups at random:HBO group and control group.Each rabbit was given a dose of MTX 100 mg/kg iv.Venous blood and cerebral spinal fluid(CSF)were taken at 1.5 h after the drug administration.A HPLC(LC 6 A)instrument was used to determine the plasma and CSF concentrations of MTX.Results:After 1.5 h of drug administration,the MTX concentration in the CSF was 2.67% of that in the plasma of the HBO group,while that of the control group was 1.24%;and significant difference was shown with paired t test( P <0 01)between the two groups.Conclusion:HBO could increase the permeability of BBB to MTX.
