环维黄杨星D对脑缺血再灌注NPY与细胞超微结构的影响

摘    要:目的:观察中药单体环维黄杨星D(CVB-D)对脑缺血-再灌注大鼠脑组织神经肽Y(NPY)与细胞超微结构的影响.方法:采用易卒中型肾血管性高血压大鼠实验,用线栓法制成一例大脑中动脉闭塞脑缺血-复流实验的实验组.实验组根据脑缺血6h后复流6、72、168h不同时间,用放免法和透射电镜分别测定上述不同时间大鼠脑组织NPY水平与细胞超微结构;观测不同时间点的脑梗塞的面积、脑含水量.结果:CVB-D能降低缺血再灌注大鼠脑内NPY的含量、脑含水量及梗塞面积.结论:CVB-D对实验性脑缺血再灌注损伤有保护作用,其机理可能与抑制NPY的释放有关.

Abstract:Subject:To observe the effect of Cyclovirobuxine D(CBV-D)on neuropeptide Y(NPY)and cell ultrastructure in brain tissue during rat cerebral ischemia and reperfusion.Methods:Stroke-prone-Renovascular-Hypertensive-Rats (RHRSP)models were estabilished by bilaterally narrowing the renal artery with silk loop clips;the experiment group of rat middle cerbral artery occlusion(MCAO)and peperfusion was made by nylon suture embolization.Neuropeptide Y and cell ultrastructure in brain tissue in experiment group were detected after 6h cerebral ischemic and reperfusion at 6h,72h and 168h by radio immunoassay and transmission electron microscope techniques.and the water content and the rate of cerebral infarction were also detected at same time.Results:CBV-D is able to reduce the content of NPY,the water content and the rate of cerebral infarction in the focal cerebral ischemia/reperfusion model.Conclusion:CBV-D has protective effects on the focal cerebral ischemia/reperfusion injury,and the inhibition of the release of NPY in the brain probably contributs to the protective effect.
