摘    要:根据人的N-乙酰化表型制定个体化剂显方案以防止药源性疾病和药物不良反应的方法已在临床上广泛应用。本文以咖啡因为探针,采用HPI。C法测定其尿排泄的代谢物5一乙酪氨基一6一甲酸氨基一3一甲基尿嘴陡(AFMU)和l一甲黄瞟吟(IX)。由于咖啡因在体内经肝脏N一乙酸基转移酶作用生物转化完全,其代谢物比值AFMU/IX可准确反映此酶的活性'"',酶活性的高低临床上表现为快或慢乙酸化表型。1实验和方法1.l仪器美国产Waters510型HPI。C色谱仪,440紫外检测器,反相已。柱(pBondapak6minX30cm)Spec·ircFlow757型UV检测器,Waters…

Abstract:The two mltabolites of caffeine, 5 - acetylamino - 6 - formylamino - 1 - mlthyhuracil (AFMU)and 1 -mlthylxanthine(IX),in urine collected from 190 health Chinese volunteers were analyzed by HPLC. The peak height ratio of AFMU/IX was used to determine the acetylators phenotypes. The results showed that 73. 68% of the subjects could be classified as fast acetylators which is similar to the ratio previousy reported for Asian with isoniazide and sulfamlthazine as the pharmacological probes. Phenotyping with caffeine as a probe via analyzing its urinary metabolites by HPLC is shown to be simple and fast. The analyzing precision is (CV% )2. 2% - 4. 4% and the rate of recovery is 94% - 95 %.
