双嘧达莫对细胞免疫的抑制作用

摘    要: 目的:观察双嘧达莫对小鼠免疫功能的影响。方法:采用小鼠末梢血白细胞计数,脾脏称重计算脾脏系数,测小鼠足垫厚度的差值确定小鼠迟发型超敏反应,用PHA和Con A 诱生的脾淋巴细胞増殖测定淋巴细胞转化功能,作为活化的小鼠脾细胞测定小鼠IL-2的活性。结果:双嘧达莫1,10,40 mg/kg抑制小鼠的迟发型超敏反应,降低小鼠末梢血白细胞计数和脾脏系数。双嘧达莫(Dip)1,10,100,1 000 μmol/L抑制PHA诱导的小鼠脾淋巴细胞转化和Con A 诱导的小鼠脾淋巴细胞增殖反应,Dip 1,10,100 μmol/L亦抑制小鼠脾细胞产生IL-2。结论:实验结果为Dip老药新用提供一定的理论依据,临床上Dip应用于一些免疫性疾病的治疗似有较大潜力。

Abstract:OBJECTIVETo observe the effects of dipyridamole (Dip) on murine immune function. METHOD:Murine peripheral white blood cells count,and spleen index, measured by spleen weighing were used to detect murine foot pad thickness difference and to define the murine delayed hypersensitivity reaction. PHA and Con A induced murine spleen lymphocyte mitogenesis were used to determine lymphocyte transformation function. The activating murine spleen cells were used to detect the activity of IL-2. RESULTS:Dip 1,10,40 mg/kg ip inhibited murine delayed hypersensitivity reaction induced by sheep red blood cells,and obviously decreased murine spleen index and peripheral white blood cells number. Dip 1,10,100,1 000 μmol/L in vitro inhibited PHA induced murine spleen lymphocyte transformation and Con A induced murine spleen T lypphocyte cells mitogenesis. Dip 1,10,100μmol/L inhibited interleukin-2 produced by murine splenocytes (1×106/ml). CONCLUSION:A theoretical basis is provided for Dip,which will have potentialities in clinical application of some immunopathy therapy.
