新蒲饮联合奥美拉唑对Hp相关性胃炎及TLR4/JAK2/STAT3通路的影响

摘    要:目的 探讨新蒲饮联合奥美拉唑对Hp相关性胃炎的疗效研究及对TLR4/JAK2/STAT3通路影响。方法 本研究将50只C57BL/6(B6)小鼠随机分成对照组(10只)和造模组(40只)。利用快速尿素酶法及吉姆萨染色证实Hp感染小鼠模型建立后,将40只造模组小鼠随机分成模型组、新蒲饮组(纯中药组)、新蒲饮联合奥美拉唑组(中西结合组)、阿莫西林 + 克拉霉素 + 奥美拉唑肠溶片组(西药组),各组小鼠分别给予相应生理盐水或药物14天。提取小鼠胃组织蛋白做免疫组化和Western Blotting,通过检测TLR4、JAK2及STAT3的表达变化情况,分析不同药物的治疗效果。结果 通过Hp连续灌胃后,吉姆萨染色发现模型组明显有Hp堆积成簇,提示造模成功。同时,本研究发现模型组相比对照组而言,TLR4、p-JAK2和p-STAT3表达均明显升高。用药后,与模型组相比,纯中药组、中西结合组和西药组Hp感染明显下降,且Western Blotting和免疫组化均提示TLR4、p-JAK2和p-STAT3表达有不同程度下降,JAK2和STAT3总蛋白在上述几组中未发现有明显改变。结论 新蒲饮联合奥美拉唑可以通过TLR4/JAK2/STAT3通路治疗Hp相关性胃炎。

Abstract:Objective To investigate the effect of Xinpuyin combined with omeprazole on Hp-associated gastritis and its effect on TLR4/JAK2/STAT3 pathway.Methods We randomly divided 50 C57BL/6 (B6) mice into control group (10) and model group (40). After the establishment of mice model of Hp infection by rapid urease method and Giemsa staining, 40 model mice were randomly divided into model group, Xinpuyin group (pure Chinese medicine group), Xinpuyin combined with omeprazole group (Chinese-Western combination group), amoxicillin + clarithromycin + omeprazole enteric-coated tablets group (Western medicine group), each group of mice were given corresponding physiological saline or drugs for 14 days. The mouse gastric tissue protein was extracted for immunohistochemistry and Western Blotting. The therapeutic effects of different drugs were analyzed by detecting the changes of TLR4, JAK2 and STAT3 expression.Results After continuous intragastric administration of Hp, Giemsa staining revealed that the model group had obvious accumulation of Hp clusters, suggesting successful modeling. At the same time, we found that the expression of TLR4, p-JAK2 and p-STAT3 was significantly increased in the model group compared with the control group. After treatment, compared with the model group, Hp infection in the pure Chinese medicine group, the Chinese-Western combination group and the western medicine group decreased significantly, and both Western Blotting and immunohistochemistry indicated that the expression of TLR4, p-JAK2 and p-STAT3 decreased to different degrees, JAK2 and No significant changes were found in the STAT3 total protein in the above groups.Conclusion Xinpuyin combined with omeprazole can treat Hp-associated gastritis through TLR4/JAK2/STAT3 pathway.
