黄连等五种中药颗粒剂对鲍曼不动杆菌生物膜的作用研究

摘    要:目的:观察黄连、黄芩、连翘、乌梅、五味子等五种中药颗粒剂对多重耐药鲍曼不动杆菌生物膜形成的影响。方法:对广东省第二中医院2018年1月~2019年4月分离的29株非重复的多重耐药鲍曼不动杆菌进行实验,并用刚果红平板法和结晶紫染色法半定量挑选生物膜阳性菌株26株;在体外建立鲍曼不动杆菌生物膜模型,通过肉汤倍比稀释法测定黄连等5种中药颗粒剂对产膜原菌最小抑膜浓度,再通过镜下观察用药前后鲍曼菌生物膜的改变。结果:不同的单体中药颗粒剂对鲍曼不动杆菌生物膜的抑制浓度有不同程度差异。黄连、乌梅、五味子对鲍曼不动杆菌生物膜抑膜浓度范围为4~8mg/mL,黄芩的抑膜浓度范围为8~16mg/mL,在实验浓度内连翘对鲍曼不动杆菌生物膜无抑膜作用。结论:黄连、黄芩、五味子、乌梅对鲍曼不动杆菌生物膜有不同程度的抑制作用,这四种中药颗粒剂体外抑制鲍曼不动杆菌的作用与其抑膜作用有关系,具体机制还需进一步研究。

Abstract:Objective: To observe the effects of five Chinese medicine granules including Coptis chinensis, Cotinus coggygria, Forsythia suspensa, Wumei and Schisandra chinensis on biofilm formation of multidrug-resistant Acinetobacter baumannii. Methods: A total of 29 non-repeated multi-drug resistant Acinetobacter baumannii isolates from Guangdong Second Traditional Chinese Medicine Hospital from January 2018 to April 2019 were tested, and biofilms were selected semi-quantitatively using Congo red plate method and crystal violet staining method. 26 strains of positive strains;Acinetobacter baumannii biofilm model was established in vitro, and the minimum inhibitory film concentration of five Chinese medicine granules such as Coptis chinensis on membraneproducing bacteria was determined by bouillon dilution method, and the abalone before and after administration was observed under a microscope Alteration of B. mannii biofilm. Results: Different monomer Chinese medicine granules had different degrees of inhibition on the concentration of Acinetobacter baumannii biofilms. The inhibitory film concentration range of Coptis chinensis, ebony plum, and Schisandra chinensis on Acinetobacter baumannii was 4~8 mg/mL, and the inhibitory film concentration range of Scutellaria baicalensis was 8~16 mg/mL. Acinetobacter biofilm has no inhibitory effect. Conclusion: Coptis chinensis, Scutellaria baicalensis, Schisandra chinensis and Wumei have different degrees of inhibition on Acinetobacter baumannii biofilm. The four kinds of traditional Chinese medicine granules inhibit Acinetobacter baumannii in vitro and its inhibitory effect. The specific mechanism needs further the study.
