天然和氧化极低密度脂蛋白对人脐静脉内皮细胞VCAM-1表达的影响

摘    要:目的 探讨天然和氧化极低密度脂蛋白(n-VLDL,ox-VLDL)对培养的人脐静脉内皮细胞(HUVEC)表达血管细胞粘附分子1(VCAM-1)的影响。方法 将n-VLDL和ox-VLDL作用于培养的HUVEC,用细胞酶联免疫吸附试验(cell ELISA)和单核细胞粘附试验检测VCAM-1蛋白在HUVEC的表达,用原位分子杂交检测VCAM-1 mRNA的表达。结果 正常培养的HUVEC表达VCAM-1,n-VLDL和ox-VLDL促进HUVEC表达VCAM-1,尤以ox-VLDL作用更强。单核细胞粘附试验显示,n-VLDL和ox-VLDL促进单核细胞向HUVEC粘附。结论 天然和氧化极低密度脂蛋白可能通过增强血管内皮细胞表达VCAM-1而促进血液单核细胞粘附于血管壁,从而在动脉粥样硬化的早期病变中起重要作用。

Abstract:Objective To investigate the effect of native and oxidized very low density lipoprotein (n-VLDL, ox-VLDL) on the expression of VCAM-1 in the cultured human umbilical vein endothelial cells (HUVEC).Methods After the HUVEC were stimulated by n-VLDL and ox-VLDL for 24 h, the expression of VCAM-1 protein in HUVEC was detected by cell enzyme linked immunosorbent assay (cell ELISA) and monocytes adhesion test. The expression of VCAM-1 mRNA was detected by in situ hybridization. Results The cultured HUVEC could express VCAM-1. Both n-VLDL and ox-VLDL could enhance the expression of VCAM-1, especially ox-VLDL. The results of monocyte adhesion test revealed that n-VLDL and ox-VLDL could enhance the adhesion of monocytes to HUVECs. Conclusion n-VLDL and ox-VLDL enhanced the recruitment of monocytes to vascular endothelial cells by stimulating the expression of VCAM-1, thus, they may play an important role in early stage of atherosclerosis.
