青秦液对高尿酸血症大鼠尿酸代谢及相关酶活性的影响

摘    要:目的观察青秦液对高尿酸血症大鼠尿酸代谢及相关酶活性的影响。方法将70只Wistar大鼠随机分为正常组、模型组、阳性药组及青秦液大、中、小剂量组。采用灌服腺嘌呤及乙胺丁醇进行造模。阳性药组按0.09mg/kg体重灌服秋水仙碱;大剂量组给予5g/(kg·d)青秦液灌胃,中剂量组给予2.5g/(kg·d)青秦液灌胃;小剂量组予1.25g/(kg·d)青秦液灌胃。采用磷钨酸法测定血尿酸和尿尿酸,采用酶比色法测定黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)的含量。结果秋水仙碱及青秦液均具有明显降低高尿酸血症大鼠血尿酸水平的作用,但二者比较差异无统计学意义(P〉0.05);在降低高尿酸血症大鼠血ADA活性作用方面,阳性药组及青秦液大、中、小剂量组均明显低于模型组(P〈0.05,P〈0.01),且青秦液中、小剂量组大鼠血ADA含量明显低于阳性药组(P〈0.05);阳性药组及青秦液大、中、小剂量组大鼠血XOD含量均明显低于模型组(P〈0.05,P〈0.01)。结论青秦液具有明显降低高尿酸血症大鼠血尿酸水平及血ADA、XOD活性的作用;在降低高尿酸血症大鼠血ADA活性作用方面,青秦液中、小剂量组明显优于阳性药组。

Effect of Qinqinye on Hyperuricemia Uric Acid Metabolism and Related Enzymes Activity

Abstract:Objective To observe the effect of Qingqinye on hyperuricemia uric acid metabolism and related enzymes activity. Methods Seventy Wistar rats were divided into normal group, model group, positive medicine group, Qingqinye high dosage group, Qingqinye middle dosage group and Qingqinye low dosage group. Model was made with adenine and ethambutol by gavage. Colchicine 0.09 mg/(kg ~ d) were administered to the rats of positive medicine group by gavage. Rats of Qingqinye high dosage group were administered Qingqinye 5 g/(kg · d), middle dosage group were 2.5 g/(kg · d) and low dosage group were 1.25 g/kg · d by gavage. The level of serum uric acid and urinary uric acid were determined by phosphotungstic acid method. The level of XOD and ADA were determined by enzymic calorimetric method. Results Colchicine and Qingqinye significantly lowered serum uric acid level of hyperuricemia rats, while there was no significant statistical difference between them (P〉0.05). Positive medicine group, Qingqinye high dosage group, Qingqinye low dosage group and Qingqinye middle dosage group reduced blood ADA activity and better than model group (P〈O.05, P〈0.01), what's more, Qingqin ye middle dosage group and Qingqinye low dosage group were obviously better than positive medicine group in this respect (P 〈 0.05). Positive medicine group, Qingqinye middle dosage group, Qingqinye low dosage group, Qingqinye high dosage group were better than model group in reducing blood XOD activity (P 〈0.05, P 〈0.01). Conclusion Qingqinye can decrease the level of serum uric acid, ADA and XOD activity in rats hyperuricemia model. Qingqinye middle dosage group and Qingqinye low dosage group were better than positive medicine group in reducing blood ADA activity.
