谷氨酰胺增强氧化应激时肺微血管内皮细胞热休克蛋白70的表达

摘    要:目的研究谷氨酰胺(Gln)对氧化应激状态下体外培养肺微血管内皮细胞(pulmonary microvascular endothelial cells,PMVEC)热休克蛋白(HSP)70表达的影响。方法采用体外培养Wistar大鼠PMVEC,分为阳性对照组(G组)、生理盐水组(PS组)、谷氨酰胺组(Gln组)、过氧化氢加生理盐水组(H2O2+PS组)和过氧化氢加谷氨酰胺组(H2O2+Gln组)。Western blotting检测PMVEC HSP70的表达情况,分别记录平均灰度值。结果与PS组(9.05±1.51)相比,Gln组(40.28±10.19)和H2O2+PS组(43.89±10.32)PMVEC HSP70表达分别增高4.5倍和5倍(P<0.01);与H2O2+PS组相比,H2O2+Gln组(65.59±14.53)HSP70表达进一步增高,约为PS组的7倍,接近G组(70.56±12.23)表达水平。结论谷氨酰胺可增强氧化应激状态下体外培养肺微血管内皮细胞HSP70表达。

Abstract:ObjectiveTo investigate the effects of glutamine on the expression of heat shock protein (HSP) 70 in pulmonary micro-vascular endothelial cells (PMVECs) under oxidative stress conditions. MethodsPMVECs from Wistar rats were divided into 5 groups: the positive control group (G group), the physiologic saline group (PS group), the glutamine group(Gln group), the H2O2+PS group and the H2O2+Gln group. Western blotting was used to investigate the expression of HSP70. ResultsCompared with the PS group, the expression of HSP70 significantly increased in the Gln group(9.05±1.51 vs 40.28±10.19) and the H2O2+PS group(9.05±1.51 vs 43.89±10.32). The expression of HSP70 was higher in the H2O2+Gln group than in the H2O2+PS group(65.59±14.53 vs 43.89±10.32). ConclusionGlutamine enhances the expression of HSP70 in pulmonary micro-vascular endothelial cells under oxidative stress conditions.
