| 摘 要： | 目的:探讨Attractin(Atrn)在培养大鼠睾丸leydig细胞的表达。方法:体外培养成年雄性SD大鼠睾丸leydig细胞,采用实时荧光定量RT-PCR检测Atrn mRNA表达,western blot检测Atrn蛋白的表达,同时应用化学发光法检测细胞睾酮分泌水平。结果:在体外培养大鼠睾丸leydig细胞过程中,Atrn mRNA、Atrn蛋白表达量和睾酮生成量逐渐减少。Atrn蛋白表达量与Atrn mRNA表达量(r=0.953,P<0.05)和睾酮生成量(r=0.976,P<0.01)成正相关。结论:大鼠睾丸Leydig细胞Atrn mRNA,Atrn蛋白表达量随培养天数的延长而减少,可能与睾酮的变化有关。

| Abstract: | Objective:To investigate the expression of Attractin(Atrn) in cultured adult rat Leydig cells.Methods:The gene and protein expression of Atrn in Leydig cells was quantitated by Real-time polymerase chain reaction(PCR) and western blot methods,respectively,while testosterone in the medium was detected by Chemiluminescence Immunoassay.Results:Expression levels of gene and protein of Atrn and the level of testosterone was decreased during culturing process.Multiple correlation analysis showed a positive correlation between the gene and protein expression levels of Atrn(r =0.953,P<0.05),and a positive correlation between the protein expression levels of Atrn and level of testosterone(r = 0.976,P<0.01).Conclusion:Expression level of Atrn is decreased in cultured Leydig cells,which may be related with testosterone production. |
