补肾活血方对SAMP8小鼠学习记忆及海马神经干细胞影响实验研究

摘    要:目的观察补肾活血方对快速老化SAMP8小鼠行为学及海马内神经干细胞的影响,以期为补肾活血方治疗阿尔茨海默病(Alzheimer’s disease,AD)的临床应用提供实验依据。方法7月龄的SAMP8雄性小鼠随机分为3组,分别为模型组、中药组和西药对照组,同月龄的SAMR1小鼠作为正常对照组。中药组按照6 g/kg补肾活血方灌胃给药,西药组给予3 mg/kg的盐酸多奈哌齐灌胃,正常组和模型组小鼠给予同体积的生理盐水灌胃。连续灌胃4周后采用水迷宫、免疫组织化学法和Western Blot法观察小鼠行为学、海马区NeuN和Nestin的表达。结果与正常老化SAMR1小鼠相比,模型组SAMP8小鼠学习记忆能力明显下降,海马组织Nestin和NeuN的表达减少(P<0.05);与模型组相比,给予补肾活血方和盐酸多奈哌齐西药干预后,SAMP8小鼠学习记忆能力有所改善,海马组织内Nestin和NeuN的表达增多(P<0.05);中药组和西药对照组SAMP8小鼠相比,学习记忆、海马组织Nestin和NeuN的表达无显著性差异(P>0.05)。结论补肾活血方通过上调SAMP8快速老化小鼠海马组织Nestin和NeuN的表达,促进神经干细胞的增殖和向神经元分化,从而改善SAMP8小鼠的学习认知记忆能力,对AD起治疗作用。

Abstract:Objective To observe the effect of Bushen Huoxue Recipe on the behavior of rapidly aging SAMP8 mice and neural stem cells in hippocampus so as to provide the experimental basis for the clinical application of Bushen Huoxue Recipe in the treatment of AD.Methods Male SAMP8 mice at the age of 7 months were randomly divided into 3 groups:model group,traditional Chinese medicine group and western medicine control group,and SAMR1 mice at the same age were taken as the normal control group.The traditional Chinese medicine group was given Bushen Huoxue Recipe with the dose of 6 g/kg.The western medicine group was given donepezidine hydrochloride with the dose of 3 mg/kg.The normal group and the model group were given the same volume of normal saline.Water maze,immunohistochemistry and western blot were used to observe the behavior of mice and the expressions of Neun and Nestin in the hippocampal area.Results Compared with normal aging SAMR1 mice,the learning and memory ability of SAMP8 mice in the model group was significantly decreased,and the expressions of NeuN and Nestin in the hippocampus were decreased(P<0.05).Compared with the model group,after the intervention of Bushen Huoxue Recipe and donepezil hydrochloride,the learning and memory ability of SAMP8 mice was improved,and the expressions of NeuN and Nestin in the hippocampus increased(P<0.05).There was no significant difference in the learning and memory as well as expressions of Nestin and NeuN in the hippocampal tissue between the traditional Chinese medicine group and the western medicine control group.Conclusion Bushen Huoxue Recipe can promote the proliferation and differentiation of neural stem cells into neurons by up-regulating the expressions of Nestin and NeuN in the hippocampal tissue of SAMP8 rapidly aging mice so as to improve the learning and memory ability of SAMP8 mice and plays a therapeutic role in AD.
