益气养阴方灌胃在白血病大鼠中的作用机制及对白血病抑制因子影响的研究

摘    要:目的探讨益气养阴方灌胃在白血病大鼠中的作用机制及对白血病抑制因子(leukemia inhibitory factor,LIF)水平的影响。方法选择2017年8月—2018年4月参与实验的SD大鼠40只作为对象,随机取SD大鼠10只,设为空白对照组。剩余30只SD大鼠采用尾静脉注射绿色荧光蛋白(FFP)-LT12细胞构建髓细胞性白血病(BNML)动物模型,建模成功后随机分为模型对照组、阿糖胞苷组及益气养阴方组,每组SD大鼠10只。模型对照组和空白对照组常规生理盐水灌胃;阿糖胞苷组建模后7 d经腹腔注射阿糖胞苷;益气养阴方组采用益气养阴方灌胃,4周干预后对各组效果进行评估,比较4组炎症因子水平、LIF水平、HE染色结果。结果益气养阴方组干预后4周炎症因子IL-2、IL-4、IL-6、IL-10、IL-17及TNF-α水平,均低于阿糖胞苷组及模型对照组(P<0.05);益气养阴方组干预后1、2、3及4周LIF水平,均低于阿糖胞苷组和模型对照组(P<0.05);HE染色结果表明:益气养阴方组和空白对照组细胞结构层次清晰,表层细胞与周围组织表面近似平行,细胞排列、形状规则,中间细胞层排列垂直于表层;模型对照组白细胞数量急剧增多,可观到核-质比异常高的巨化细胞;阿糖胞苷组HE染色组织结构出现紊乱,但是多数细胞正常,仅出现零星的白细胞。结论益气养阴方灌胃用于白血病大鼠中能降低炎症因子水平,有助于改善LIF水平,减轻白细胞组织浸润,能为白血病治疗提供新的思路与方法。

Abstract:Objective To investigate the action mechanism of Yiqi Yangyin Recipe for intragastric administration on leukemia rats and its effect on levels of leukemia inhibitory factor(LIF).Methods A total of 40 Sprague-Dawley rats were enrolled in the study from August 2017 to April 2018.A total of 10 SD rats were randomly selected and used as a blank control group.The remaining 30 SD rats were injected with green fluorescent protein(FFP)-LT12 cells into the tail vein to construct a myeloid leukemia(BNML)animal model.After successful modeling,they were randomly divided into model control group,cytarabine group and Yiqi Yangyin Recipe group,10 SD rats in each group.The model control group and the blank control group were given normal saline.The cytarabine group was intraperitoneally injected with cytarabine 7 days after modeling.The Yiqi Yangyin Recipe group was treated with Yiqi Yangyin Recipe for intragastric administration.After intervention for 4 weeks,the effects of each group were evaluated,and the levels of inflammatory factors,LIF levels and HE staining were compared.Results The levels of inflammatory factors IL-2,IL-4,IL-6,IL-10,IL-17 and TNF-αin the Yiqi Yangyin Recipe group were lower than those in the cytarabine group and model control group(P<0.05).LIF levels at 1,2,3 and 4 weeks after intervention in Yiqi Yangyin Recipe group were lower than those in cytarabine group and model control group(P<0.05).HE staining results showed that the cell structure of Yiqi Yangyin Recipe group and blank control group was clear,the surface cells were almost parallel to the surrounding tissue surface,the cells were arranged and the shape was regular,and the middle cell layer was arranged perpendicular to the surface layer.The number of white blood cells in the model control group increased sharply.Giant cells had abnormally high nuclear-mass ratio.HE staining of cytarabine group was disordered,but most cells were normal,and only sporadic white blood cells appeared.Conclusion Yiqi Yangyin Recipe can reduce the levels of inflammatory factors in leukemia rats,improve the level of LIF,reduce the infiltration of leukocyte tissue,and provide new ideas and methods for leukemia treatment.
