摘    要:目的采用高效液相色谱法建立鹿茸中核苷类成分指纹图谱,进行聚类分析与主成分分析,并比较梅花鹿鹿茸药材中6种核苷成分的差异。方法采用高效液相色谱法对15批不同产地梅花鹿鹿茸进行测定,以甲醇-0.07%冰醋酸为流动相梯度洗脱,流速1 mL·min^-1,检测波长为254 nm。运用国家药典委员会"中药色谱指纹图谱相似度评价软件"2012版进行评价,并对6种核苷类成分进行含量测定。结果建立了鹿茸核苷类成分HPLC指纹图谱,确定了6个共有峰构成鹿茸药材的特征峰,且方法学考察达到标准。结论该法所建立的鹿茸核苷类指纹图谱特征性强、方法简便,结合6种核苷类成分含量测定可更好控制其质量,对梅花鹿鹿茸的鉴定及质量控制具有指导意义和参考价值。

HPLC Fingerprints Integrated with Quantitative Determination of Multi-components for Quality Control of Pilos Antler

Abstract:Objective The HPLC-DAD method was used to establish the fingerprints of nucleosides in pilos antler and carry on the perform cluster analysis and principal component analysis and compare the differences of six nucleosides in sika deer antler. Method Fifteen batches of sika deer antlers from different origins were determined by HPLC and linear gradient elution was Methanol-0.07% glacial acetic acid,with the flow rate of 1 mL·min^-1 and detection wavelength at 254 nm. We assessed 15 batches from different origins in China by Chromatographic Fingerprint of Traditional Chinese Medicine(2012) and determined the contents of six principal components.Result The HPLC fingerprints of antler nucleosides were established, and the six common peaks were determined to constitute the characteristic peaks of velvet medicinal materials, and the methodological investigation reached the standard. Conclusion This study indicated that the combination of quantitative and chromatographic fingerprint analysis can be readily utilized as a quality control method for pilos antler.
