摘    要:目的探究白芍-柴胡药对治疗肝细胞癌(hepatocellular carcinoma,HCC)的有效活性化合物及其靶点的作用机制。方法本研究通过TCMSP数据库筛选出白芍-柴胡药对中的化合物以及相关靶点蛋白,并运用Uniprot数据库和Genecard数据库查询与靶点蛋白相对应的人类基因名称,进行标准化处理;通过Genecards数据库挖掘肝细胞癌疾病靶点,将其与中药靶点蛋白映射;将映射出的药对治疗HCC的有效靶点导入Cytoscape 3.2.1构建网络图,并运用CTD数据库进行GO富集和KEGG通路富集。结果获得白芍-柴胡药对治疗HCC的共22个成分和170个有效靶点;获得16条GO生物过程和7条相关信号通路,涉及代谢通路(Metabolic pathways)、Ras通路(Ras signaling pathway)、PI3K-Akt信号通路(PI3K-Akt signaling pathway)等。结论白芍-柴胡药对活性靶点主要通过代谢通路、Ras信号通路、PI3K-Akt信号通路调控肝癌细胞过程及其代谢来发挥治疗作用,为后续实验提供了思路与依据。

Abstract:Objective This study was designed to explore the possible mechanisms of components and targets of Radix paeoniae alba and Radix bupleuri in the treatment of hepatocellular carcinoma(HCC)based on the network pharmacology.Methods The active components and targets of Radix paeoniae alba and Radix bupleuri were obtained through TCMSP.By using Uniprot database and GeneCards database,the genes of Radix paeoniae alba and Radix bupleuri were collected.By using genecards database to collect relatively targets of HCC.And we mapped them with the TCM targets,constructing the"components-targets"network of and"targets-pathways"network with Cytospace 3.6.1 software.The GO enrichment and the KEGG enrichment of targets were completed by CTD database.Results Twenty-two active components including kaempferol,beta-sitosterol,quercetin,isorhamnetin,Stigmasterol and 170 active targets crucially including VEGF,NF-κB,CDKs,PTGS1 and PTGS2 were collected.Sixteen GO biological pathways and seven related signaling pathways,involving metabolic pathways,Ras signaling pathwaysand phosphatidylinositol 3-kinase(PI3 K)/Akt signaling pathways.Conclusion The therapeutic mechanism ofRadix paeoniae alba and Radix bupleuriactive targets in treating HCC may involve regulating the cellular process and metabolic process based on metabolic pathways,Ras signaling pathway and PI3 K-Akt signaling pathway.This consequence may provide a direction for further researches.
