津力达颗粒对PCOS模型大鼠卵巢TGF-β/CTGF的影响

摘    要:目的:研究津力达颗粒对多囊卵巢综合征(PCOS)大鼠卵巢TGF-β1/CTGF表达的影响。方法:40只21 d龄雌性SD大鼠皮下注射脱氢表雄酮(DHEA)建模,按随机数字表法分为模型组、二甲双胍组、津力达组和津力达+二甲双胍组,各组分别给予相应药物灌胃,另取10只空白组皮下注射大豆油,模型组给予等量生理盐水灌胃。实验结束后,观察卵巢病理学变化,检测各组大鼠TGF-β1、CTGF血清学水平、卵巢蛋白含量以及mRNA表达。结果:模型组较空白组血清TGF-β1、CTGF水平及卵巢TGF-β1、CTGF蛋白含量及mRNA显著增高,差异有统计学意义;与模型组比较,二甲双胍组、津力达组和津力达+二甲双胍组TGF-β1、CTGF蛋白及mRNA表达均有下降,差异有统计学意义,表明当津力达与二甲双胍联合应用时下降趋势增加。结论:津力颗粒可能通过调节卵巢组织中TGF-β1/CTGF表达,改善PCOS模型大鼠卵巢纤维化水平。

Abstract:Objective: To investigate the effects of Jinlida Granule on the expression of TGF-β1/CTGF in the ovary of rats with polycystic ovary syndrome(PCOS), and to explore the mechanism of action of Jinlida in the treatment of PCOS. Methods:To establish PCOS rat model,21-days old female SD rats were injected subcutaneously with 6 mg/100 g weight of DHEA per day plus 0.2 mL injection of soybean oil for 20 days, while rats in control group were injected with the same volume of oil. The PCOS rats were randomly divided into model group, metformin group, Jinlida group and Jinlida+metformin group. Each group includs 10 PCOS rats. After the PCOS rat model was established, each group was given different drugs interventions for 28 days. After the experiment, the pathological changes of ovary were observed under light microscope. The serum levels of TGF-β1, CTGF, ovarian protein and mRNA expression were detected by ELISA, immunohistochemistry, Western blot and RT-PCR in each group of rats. Results: The levels of serum TGF-β1, CTGF and ovarian TGF-β1, CTGF protein content and mRNA were significantly higher in the model group than in the blank group. Compared with the model group, the metformin group, the Jinlida group and the Jinlida+Metformin group TGF-β1, CTGF protein and mRNA expression decreased with statistical difference. When Jinlida was combined with metformin, the decreasing trend increased. Conclusions: Jinlida Granule may improve the ovarian fibrosis level in PCOS model rats by regulating the expression of TGF-β1/CTGF in ovarian tissue.
