罗汉果黄酮干预运动大鼠骨骼肌FLK-1及BFGF表达的影响

摘    要:目的:探讨补充罗汉果黄酮对训练大鼠腓肠肌FLK-1及BFGF mRNA及蛋白表达的影响。方法:将50只大鼠随机分为安静对照组、运动对照组、运动+低、中、高剂量罗汉果黄酮组,各训练组大鼠进行为期6周的力竭游泳训练并给予低、中、高剂量组不同剂量的罗汉果黄酮后(给药6周,每周6 d),采用RT-PCR和Western Blot法测定大鼠腓肠肌FLK-1及BFGF mRNA及蛋白表达。结果:腓肠肌ME组FLK-1 mRNA及蛋白表达均高于NC组,腓肠肌MGFM和MGFH组FLK-1 mRNA表达高于ME组,腓肠肌MGFH组FLK-1蛋白表达高于ME组,腓肠肌ME组BFGF mRNA及蛋白表达均高于NC组,MGFH组BFGF mRNA表达高于ME组,MGFM和MGFH组BFGF蛋白表达高于ME组。结论:力竭训练引起大鼠腓肠肌FLK-1及BFGF mRNA及蛋白表达存在差异性,补充罗汉果黄酮增加了肌肉组织供血能力和提高氧运输能力,从而有利于提高大鼠的运动能力。

Abstract:Objective:To investigate the effect of mogrosides flavonoids on the expression of FLK-1 and BFGF mRNA and protein in the gastrocnemius muscle of trained rats.Methods:50 rats were randomly divided into the control group,exercised control group,exercised+low dose,medium dose and high dose mangosteen flavonoids groups.The rats in each training group were subjected to exhaustive swimming training for 6 weeks.The doses of low,medium and high dose of mogroside flavonoids,The gastric perfusion was administered for 6 weeks,and 6 days/week.RT-PCR method and Western Blot methods were used to determined the mRNA and protein expressions of FLK-1 and BFGF in the rat gastrocnemius muscle.Results:The expression of FLK-1 mRNA and protein in the gastrocnemius muscle ME group was higher than that in the NC group.The expression of FLK-1 mRNA in the gastrocnemius muscle MGFM and MGFH groups was higher than that in the ME group.The expression of FLK-1 protein in the gastrocnemius muscle MGFH group was higher than that in the ME group.The expression of BFGF mRNA and protein in the gastrocnemius muscle ME group was higher than that in the NC group.The expression of BFGF mRNA in the MGFH group was higher than that in the ME group.The expression of BFGF protein in the MGFM and MGFH groups was higher than that in the ME group.Conclusions:The expression of different FLK-1 and BFGF mRNA and protein in the gastrocnemius of rats by exhaustive training,Supplementing Mangosteen flavone to increase muscle tissue blood supply capacity and improve oxygen transport capacity,is conducive to improve athletic ability.Exhaustive training caused differences in the expression of FLK-1 and BFGF mRNA and protein in rat gastrocnemius muscle.And supplementing Mogrosides Flavonoids increased muscle tissue blood supply capacity and increased oxygen transport capacity,which could be beneficial to improve the exercise capacity of rats.
