基于网络药理学研究加味桃核承气汤治疗2型糖尿病的作用机制

摘    要:目的:基于网络药理学方法探讨加味桃核承气汤(jiawei taohe chengqi tang,JTCT)治疗2型糖尿病(type 2 diabetes mellitus,T2DM)的作用机制。方法:通过TCMSP获得JTCT药物成分,预测其潜在靶点并于OMIM、TTD、DiGSeE等5个疾病靶点数据库进行比对,获得JTCT治疗T2DM相关靶点。通过String和Cytoscape构建PPI网络,筛选degree排名前5的靶点。采用Metascape对JTCT治疗T2DM相关靶点进行KEGG分析,通过Cytoscape构建JTCT治疗T2DM“成分-靶点-通路”网络。结果:共筛选出112个成分45个相关靶点,包括AKT1、SRC、EGFR等,PPI网络中degree排名前5的靶点分别为AKT1、SRC、EGFR、CASP3、NOS3。经过KEGG分析,JTCT治疗T2DM参与雌激素信号通路、VEGF信号通路、AGE-RAGE信号通路等通路。结论:本研究初步预测JTCT治疗T2DM的作用机制,为进一步临床应用提供了理论依据。

Abstract:Objective:To study the mechanism of Jiawei Taohe Chengqi Decoction(JTCD)in the treatment of type 2 diabetes mellitus(T2DM)based on network pharmacology.Methods:JTCD drug components were obtained by TCMSP,and their potential targets were predicted and compared in five disease target databases such as OMIM,TTD and DiGSeE,and JTCD was used to treat T2DM related targets.The PPI network was constructed by String and Cytoscape,and the top 5 targets of degree were screened.KEGG analysis of JTCD-treated T2DM-related targets was performed using Metascape,and JTCD was constructed by Cytoscape to build the T2DM"component-target-path"network.Results:A total of 112 components and 45 related targets were screened,including AKT1,SRC,and EGFR.The top 5 targets in the PPI network are AKT1,SRC,EGFR,CASP3,and NOS3.The top 5 targets in the PPI network are AKT1,SRC,EGFR,CASP3,and NOS3.After KEGG analysis,JTCD treatment of T2DM involved in estrogen signaling pathway,VEGF signaling pathway,AGE-RAGE signaling pathway and other pathways.Conclusions:This study preliminarily predicted the mechanism of action of JTCD in the treatment of T2DM,and provided a theoretical basis for further clinical application.
