亚低温对大鼠颅脑外伤后脑组织AQP-4和Caspase-3表达的影响

摘    要:目的观察亚低温治疗对实验大鼠颅脑外伤后水通道蛋白4(AQP-4)和半胱天冬酶3(caspase-3)的影响,探讨亚低温对颅脑外伤后可能的脑保护分子生物机制。方法健康成年雄性Wistar大鼠45只,采用Feeney法建立脑外伤动物模型,随机分为对照组、创伤组和亚低温组,每组15只,亚低温组在损伤后立即给予亚低温暴露,用干湿重法测定脑组织含水量、免疫组织化学法及图象分析技术检测AQP-4和caspase-3及双重染色免疫组织化学法检测AQP-4和S-100。结果与假手术组比较,大鼠颅脑外伤后脑组织AQP-4和caspase-3的表达及脑组织含水量明显升高(P0.05);亚低温治疗后,大鼠脑组织AQP-4和caspase-3的表达及脑组织含水量较脑创伤组明显降低(P0.05)。结论亚低温对创伤性脑损伤的脑保护机制可能与减轻胶质细胞介导的脑水肿,减少神经细胞凋亡有关。

Abstract:Objective The effect of mild hypothermia on the expression of aquaporin-4 and caspase-3 after brain injury in rats is observed and the neuroprotective mechanism of mild hypothermia on brain injury is discussed.Methods Forty-five adult healthy male Wistar rats were used in the study.The animal models were established by Feeney method,and all the rats were randomly divided into three groups:control group (15 cases),traumatic group (15 cases) and mild hypothermia group (15 cases).The changes of brain water content (BWC) were measured by the wet and dry weight methods.Immunohistochemistry was used to determine the changes of expression of AQP4 and caspase-3 protein,and double dyed immunohistochemistry was used to determine AQP4 and S100.Results Compared with control group,the expressions of AQP4,caspase-3 and BWC were obviously higher in traumatic group; but after mild hypothermia treatment,the expressions of AQP4,caspase-3 and BWC were obviously lower compared with traumatic group.Conclusion The protection of mild hypothermia on brain trauma may be associated with the amelioration of neuroglia cell-mediated brain edema and the decrease of apoptosis of neural cells.
