牛蒡子改善糖尿病大鼠肾脏病变机制的探讨

摘    要:目的探讨牛蒡子对糖尿病大鼠肾脏保护作用及其可能机制。方法腹腔内注射链脲佐菌素制备糖尿病模型,分析各组大鼠肾组织NF-κB、单核细胞趋化蛋白-1(MCP-1)、纤粘蛋白(FN)的表达,并分析NF-κB与24h尿蛋白及血肌酐相关性。结果糖尿病组肾小球中NF-κB、MCP-1和FN表达高于对照组(P〈L0.01)。与糖尿病组相比,牛蒡子明显抑制NF-κB活化,降低MCP-1、FN表达(P〈0.05),减少24h尿蛋白排泄(P,0.01),改善肾功能指标及肾组织病理学损害。结论牛蒡子对糖尿病大鼠肾脏病变具有明显防治作用。抑制NF-κB的活化可能是牛蒡子发挥肾脏保护作用的机制之一。

Abstract:OBJECTIVE To observe the effect of Fructus Arctii on diabetic nephropathy (DN) m expenmental diabetic rats and to study its mechanism. METHODS Diabetic model was made by intraperitoneal injection of streptozocin. Expressions of NF-κB, Monocyte Chemo-attractant Protein-1 (MCP-1) and fibronectin (FN) in glomeruli,24-hour urine protein, triglyceride (TG), total cholesterol (TC) and plasma creatinine (Cr) were mea- sured. RESULTS Activity of NF-κB in renal tissues, expression of FN and MCP-1 in glomeruli in diabetic rats were significantly higher than those in normal rats (P 〈 0.01 ). Fructus Arctii treatment significantly inhibited activation of NF-κB (P 〈 0.05), reduced expression of MCP-1 and FN, decreased urine protein excretion and levels of TG and TC, improved renal function, suppressed glomerular enlargement, mesangial cells proliferation and extracellular matrix accumulation, compared with diabetic rats. CONCLUSION These results suggested that Fructus Arctii treatment slowed down the development of DN. Renoprotective effect of Fructus Arctii on DN may be mediated throng inhibition of activation of NF-κB.
