摘    要:目的研究四氢呋喃(tetrahydrofuran,THF)与牛血清白蛋白(bovine serum albumin,BSA)相互作用的机制,探讨温度对其反应的影响。方法移取的BSA于石英比色皿中,搅拌混合均匀,避光静置,分别在25℃(298 K)、29℃(302 K)、33℃(306 K)、37℃(310 K)下以280 nm为激发波长,在LS-55荧光分光光度计上扫描280~500 nm波长范围内的发射光谱。结果不同温度下的结合猝灭常数(Ksv)为(3.466~6.317)×104L/mol,猝灭常数随温度的升高而减小。结论四氢呋喃对BSA的猝灭作用是属于静态猝灭。

Abstract:[Objective]To study the mechanism of the interaction between tetrahydrofuran( THF) and bovine serum albumin( BSA),probe the influence on the action caused by temperature.[Methods]BSA was moved into quartz colorimetric dish,mixed with a certain amount of THF,and let stand in dark condition. The emission spectrum in 280 ~500 nm wavelength range was scanned using the LS-55 fluorescence spectrophotometer at 25 ℃( 298 K),29 ℃( 302 K),33 ℃( 306 K) and 37 ℃( 310 K) with the excitation wavelength of 280 nm.[Results]The quenching constants( Ksv) at different temperature were between 3. 466 ×104L/mol and 6. 317 ×104L/mol,and decreased with increasing temperature.[Conclusion]The fluorescence quenching of BSA by THF is identified as the static quenching.
