神经生长因子在特应性皮炎小鼠模型Th1/Th2类细胞因子免疫失衡中的作用

摘    要:目的探讨神经生长因子(NGF)在特应性皮炎小鼠Th1/Th2类细胞因子免疫失衡中的作用。方法将雌性BALB/c小鼠随机分为四组:正常对照组、特应性皮炎组(AD组)、NGF干预组(NGF组)及抗NGF抗体干预组(NGF阻断组),每组6只。用卵清蛋白经皮肤致敏建立小鼠AD模型。ELISA测定血清中总IgE水平,RT-PCR半定量检测皮肤组织中IL-4和IFN-γmRNA的表达。结果NGF处理组中血清总IgE水平和皮肤组织IL-4mRNA表达水平均高于未处理AD组[(7292.154±1352.654)pg/mL和(4782.145±1132.121)pg/mL;(78.50±3.94)%和(60.67±3.56)%](P<0.01),而IFN-γmRNA表达呈现反相变化[(24.33±2.94)%和(43.17±5.46)%](P<0.01)。NGF抗体干预处理,能阻断NGF诱导IgE和IL-4mRNA的表达[(1475.532±512.476)pg/mL;(39.83±2.64)%](P<0.01)。结论NGF通过诱导Th1型向Th2型免疫应答偏移来参与特应性皮炎Th1/Th2类细胞因子免疫失衡效应。

Abstract:Objective To explore the effect of nerve growth factor(NGF)on Th1/Th2 cell immune imbalance in atopic dermatitis mice.Methods 24 female BALB/c mice were divided into 4 groups:normal control group,atopic dermatitis(AD) group,NGF group,and anti-NGF-antibody group.The mouse model of atopic dermatitis were established by sensitization with ovalbumin.Serum IgE level was measured by ELISA.Expression of IL-4 and IFN-γmRNA was detected by RT-PCR.Results As compared with AD group,the level of serum total IgE and the expression of IL-4 mRNA in skin tissue were enhanced remarkedly in the NGF group[(7292.154±1352.654) pg/mL vs (4782.145±1132.121) pg/mL;(78.50±3.94)% vs (60.67±3.56)%](P<0.01),however the expression of IFN-γmRNA was decreased significantly [(24.33±2.94)% vs (43.17±5.46)%](P<0.01).When the activity of NGF was blocked by anti-NGF antibody,the levels of serum total IgE and IL-4 mRNA in skin tissue were repressed[(1475.532±512.476)pg/mL;(39.83±2.64)%](P<0.01).Conclusion Through induction of Th1 type to Th2 type immune response deviation,NGF may be involved in Th1/Th2 immune imbalance in the atopic dermatitis.
