半夏遗传多样性的RAPD分析

摘    要:目的对半夏的遗传多样性、亲缘关系进行研究。方法采用RAPD分子标记技术,使用80个随机引物对其遗传多样性进行了研究。结果半夏物种水平上的多态性比率为88.06%,其中野生半夏(88.06%)比栽培半夏(84.31%)要高。在群体水平上,野生半夏多态位点比率的加权平均值(82.61%)比栽培半夏(84.31%)略低。结论半夏由野生转栽培对其遗传多样性影响不大,各个产地半夏种内遗传差异小;但钟祥野生半夏多态位点数最高,遗传信息最丰富,质量优。

Analysis of Genetic Diversity in Pinellia ternate by RAPD
Abstract:Objective To study genetic relationship and diversity of P. ternate species from different regions. Methods By the method of random amplified polymorphin DNA( RAPD), the genetic diversity and sibling relationship of above species were studies by 80 RAPD primers. Results The percentage of polymorphic bands of P. ternate is 88.06%, and the wild rate (88.06% ) is higher than that of cultured( 84.31%)at the level of its speccies. While the average percentage of loci per primer of wild is 82.61%, which is a little lower than the cultured (84.31%) at the level of its populations. Conclusions It had the highest polymorphic bands and loci per primer from P. ternate in Zhongxiang county of Hubei province, which showed that it has the richest genetic information and highest quality. It has no significant influence on the genetic diversity among each regions and between the wild and cultured. And it had some sibling relationship from P.ternate in Jingzhou county of Hubei province with other regions, it was showed that P. ternate from Jingzhou county probably cultured or nearly cultured form the other regions.
