甘草甜素对接触性皮炎小鼠表皮内结合珠蛋白的影响

摘    要:目的探讨甘草甜素对接触性皮炎小鼠表皮内结合珠蛋白的影响。方法 126只接触性皮炎小鼠分别给予生理盐水、不同剂量的甘草甜素及地塞米松。采用RT-PCR和原位杂交的方法检测接触性皮炎小鼠表皮内结合珠蛋白的表达。结果甘草甜素5 mg/kg和10 mg/kg组,激发后24和48 h表皮内结合珠蛋白表达增强,甘草甜素80 mg/kg组和地塞米松1 mg/kg组,激发后24和48 h表皮内结合珠蛋白表达减弱或消失。结论低剂量甘草甜素能增强接触性皮炎小鼠表皮内结合珠蛋白的表达,而高剂量甘草甜素则减弱结合珠蛋白的表达。

Abstract:Objective To investigate the effect of glycyrrhizin(GL) on haptoglobin(Hp)mRNA expression in mice epidermis of contact dermatitis.Methods 126 mice of contact dermatitis were treated with normal saline,different doses of GL and dexamethasone respectively.The Hp mRNA expression was detected by RT-PCR and in situ hybridization.Results The expression of Hp mRNA in groups treated with GL 5 and 10 mg/kg were increased more significantly than those of normal saline at 24 and 48 h after challenging.The expression of Hp mRNA in groups treated with GL 80 mg/kg and dexamethasone 1 mg/kg were decreased more significantly than those of normal saline at 24 and 48 h after challenging.Conclusion The expression of Hp mRNA in mice epidermis of contact dermatitis can be regulated bidirectionally by GL.The expressions of Hp mRNA may be increased by lower dose,but decreased by higher dose of GL treatment.
