摘    要:目的:提取茸毛链球菌主要表面蛋白抗原(PAg)对其性质进行初步鉴定。方法:用硫酸铵沉淀、DEAE-cel-lulose离子交换和 Sepharose 6B凝胶过滤从茸毛链球菌 6715培养上清中提取 PAg,并对其分子量,蛋白质含量,氨基酸组成及免疫学特性进行检测。结果与结论:PAg能与特异性抗血清发生反应,SDS-PAGE呈单一区带,分子量为210KD,蛋白质含量为61.4%,氨基酸组成分析揭示其酸性和碱性氨基酸分别为26.7%和10.7%。

Abstract:Objective: To purify and characterize the major cell surface protein antigen of streptococcus sobrinus. Methods: PAg was purified by 65 % saturate ammonium sulfate precipitation, DEAE - cellulose ion - exchange chromatography and Sepharose 6B gel - filtration chromatography. Results and Conclusion: SDS-PAGE showed the extract PAg was pure and in the ranges of 210KD, protein quantitative analysis showed PAg contained 61.4 % proteins, amino acid analysis revealed that PAg contained 26. 7 % acidic and 10.7 % basic acid residues.
