摘    要:用神经细胞体外培养技术,在光、电镜下进行定量组织化学、免疫组化方法观察17—18天大鼠胚胎下丘脑神经元体外生长发育过程。结果显示:细胞培养3天时,神经元以2个突起为主,占81%;1周时,神经元以3个突起为主,占51%;2周时神经元以3~4个突起为主,分别占37%和31%。定量组化结果显示神经元SDH、AChE含量随培养时间延长而增多,在第3天分别为64.2和3.8,1周时分别为98.2和12.8,2周时分别为127.6和24.6。免疫组化结果显示培养神经元在第3天时,神经元特异烯醇酶(NSE)呈阳性反应。上述结果提示,体外培养胚胎大鼠下丘脑神经元在第3天基本发育成熟。本文还对体外培养神经元的特点进行了详细的描述。

OBSERVATION ON MORPHOGENESIS OF HYPOTHALAMIC NEURONS OF EMBRYO RAT IN VITRO
Abstract:Hypothalamic neurons of 17-18 day embryo rat were cultured and observed by light and electron microscopy combined with immunohistochemistry or quantitative hjstochemistry. It was shown that culture neurons gave rise to 2 processes (in 81%) on day 3, 3 processes (in 51%) on day 7, and had 3 and 4 processes(in 37% and 31%, respectively) by the end of 2 weeks, Histochemical study showed. that the concentration of SDH and AChE in the neuron was increasing during culture. The concentrations Were 64,2 and 3.8, 98.2 and 12.8, and 127.6 and 24.6 on day 3, day 7and:day 14 respe ctively, Immunohistochemistry showed that NSE began to be positive on day 3. The results suggested that the hypothalamic neurons of 17—18 day rat embryo began to mature on day 3 of culture.
